Measuring Gene Flow in the Cultivation of Bt Brinjal (2016--2017)

Abstract/ Executive Summary:

Bt brinjal was introduced in Bangladesh in 2013 as first genetically engineered crop. The inevitable coexistence between transgenic and conventional Begun requires the assessment of transgene outflow. The study was carried out at Biotechnology Research field, BARI, Gazipur during the winter season of 2016-17. The experimental design consisted of a Bt plot surrounded by non-transgenic plants. Different distances i.e. 1m, 2m, 4m, 6m, 9m and 18m between the Bt and the non-Bt plants at four directions (North, South, East and West) were considered. Standard cultural management practices were applied in both Bt and non-Bt brinjal plants. After harvesting of non-Bt fruits, the presence of transgene (Cry1Ac) was assessed lateral flow strip method using DGX020-DesiGenXpresstrips Kit (Mahyco, India). On an average of four directions, the plants showed outcrossed at the distance of 1m, 2m, 4 m, 6m, 9m and 18m were 14%, 8.5%, 3.5%, 2.25, 2.5% and 1.75%, respectively. In total, 130 out of 2400 plants (5.42%) showed outcrossed. The study should be continued in the next season for further confirmation

Objectives:

As biosafety measures, it is necessary to study the gene flow on Bt gene from Bt brinjal to non-Bt brinjal plants under Bangladesh context.

Methodology:

The experiment was carried out in the Biotechnology Research field, BARI, Gazipur during 2016-17. A 12 m x 12m block of BARI Bt Begun 2 was planted at the center of the experimental field. The central block containing Bt brinjal plants was surrounded by its counterparts non- Bt i.e. BARI Begun 4 (Kazla) plants at 1, 2, 4, 6, 9 and 18 m distance at 4 directions (north, south, east, and west). Seeds were harvested from the mature and ripened non-Bt fruits. The lateral flow strip method was followed to test the presence of Cry1Ac gene in the non-Bt seedlings. Wind speed data were collected from the Plant Physiology Division, BRRI, Gazipur.

Achievement/Findings:

The distance was the key factor that affected Cry1Ac gene flow by pollen dispersal. The proportion of seeds containing the Cry1Acgene decreases with the increasing distance away from the transgenic Bt plants. Most of the gene flow was observed 1m away from the Bt brinjal plants, about 8-21% with an average of 14%. When the distance from the transgenic plants increased to 2 m, 8.5% of plants exhibited cross-pollination. When the distance was increased to 4 m, 6m, 9m and 18m, the plants showed outcrossed as 3.5%, 2.25, 2.5% and 1.75%, respectively. On average, 130 plants out of 2400 plants (5.42%) showed outcrossed. Brinjal is diploid (2n=24) in nature with a nuclear genome content of 1100Mbp. Hence, it may be categorized as often cross-pollinated species. Mahyco (2009) found the maximum distance the pollen from the central transgenic brinjal plot traveled was to be 30 meters, and an average of 87 progeny plants out of 13600 showed out-crossing, giving an out-crossing percentage of 0.64% over the two locations in India.
