In Vitro Regeneration of Bitter Gourd (momordica Charantia L.) (2017--2018)

Abstract/ Executive Summary:

For the development of a suitable regeneration protocol of bitter gourd, leaf segments of BARI Korala-1 and Tia were cultured on MS medium supplemented with different concentrations of plant growth regulators. The types of responses were different in various plant growth regulators. Among the treatments 1.5 mg/l and 2.0 mg/l concentrations of 2,4-D treated media found better for callus formation.

Objectives:

A regeneration protocol is a prerequisite for genetic transformation technology. Many factors are responsible for in vitro regeneration which needs to be standardized.

Methodology:

BARI Korala-1 seeds were collected from Horticulture Research Centre, Bangladesh Agricultural Research Institute, Gazipur and ‘Tia’ was collected from Lal Teer Seed Company. Seeds were sown in the soil. After germination, young leaves were sterilized with Clorox solution (60 ml Clorox: 40 ml distilled water) with 2 drops of Tween-20 for 5-6min followed by 3-4 times washing with sterile distilled water than 70 % Ethanol for 30 seconds followed by 3-4 times washing with sterile distilled water inside the laminar airflow cabinet. The explants were cut off to leave 2-5 mm. The prepared explants were placed in MS media (Murashige and Skoog, 1962) with different concentrations and combinations of 2, 4-D. All cultures were incubated at 25 ± 1⁰C with a 16 h photoperiod (2000 lux) provided by cool white fluorescent tubes for three weeks.

Achievement/Findings:

There was a significant effect of 2, 4-D on the callus formation of BARI Korola-1. The highest percent (100%) explant produced callus in T3 and T4 treatments of 2, 4-D treated media of BARI Korola-1. Callus quality was also found very well in the two treatments. On the other hand, 95% explant produced callus in T5 treatment and the lowest from T₁ treatment (80%). Fresh weight was increased with the increasing of 2, 4-D up to T4 then after it decreased. A similar trend was found in dry weight. Effect of 2, 4-D on the frequency of callus formation from the leaf of ‘Tia’ .A higher percentage of callus was found from T₃ treatment (95%) followed by T4 and T5 treatment (90%). The comparatively lower percentage of callus was obtained from T1(70%). Fresh weight and dry weight were higher with the application of T3 in MS media. The colour of the calli was yellowish to greenish. The maximum morphogenic callus from leaf explant by culturing in MS medium supplemented with 2.5 mg/l 2, 4-D + 3.0 mg/l BAP. Callus was transferred into the different concentrations of BAP and AgNO_3,but did not produce the shoot. Malik et al. (2007) also did not found any shoots from different explants of bitter gourd. Different combinations of shooting media are being induced to get shoots from the calli. Among the treatments, T3 and T4 treatments of 2,4-D and TDZ treated media found better for callus formation.
