In Vitro Regeneration of Grass Pea (lathyrus Sativus L.) (2017--2018)

Abstract/ Executive Summary:

Internode explants of BARI Khesari-3 were cultured in vitro on MS medium supplemented with combinations of varying concentrations of thidiazuron (TDZ), 6-benzylaminopurine (BAP) with napthalic acetic acid (NAA) combinations, zeatin and indole acetic acid IAA combinations, TDZ and NAA combination along with a control treatment without any growth regulators. Only callus formation was observed from cultured explants. The highest percentage (75%) of response with the best callus growth was observed in 2.0 mg/l TDZ + 0.1 mg/l and occasional shoot induction was observed in 1.0 mg/l BAP + 0.1 mg/l media.

Objectives:

This experiment was undertaken to develop and standardize an efficient regeneration protocol of some BARI-released varieties of grass pea.

Methodology:

BARI Khesari-3 seeds were collected from Pulse Research Centre, Bangladesh Agricultural Research Institute. Clean and healthy seeds were washed with 70% ethanol and sterilized for 25 minutes in 60% (v/v) sodium hypochlorite containing 3-4 drops Tween 20 in a laminar airflow cabinet. Afterwards, they were rinsed 4 times with sterile distilled water and transferred to ½ strength MS medium (Murashige and Skoog, 1962) and kept in dark for germination. Internodes from 1-week old seedlings were excised and cultured on MS medium supplemented with varying concentrations of thidiazuron (TDZ) (T₁, T₂, T₃ and T₄); 6-benzylaminopurine (BAP) and Napthalic acetic acid (NAA) combinations (T₅, T₆, T₇ and T₈); Zeatin and Indole acetic acid IAA combinations (T₉, and T₁₀); TDZ and NAA combination (T₁₁) along with a control treatment T₀ without any growth regulators (MS). All cultures were incubated at 25 ± 1⁰C with a 16 h photoperiod (2000 lux) provided by cool white fluorescent tubes. The pH of the medium was adjusted to 5.8. Explants were subcultured every 14 days on the same media and kept under the same conditions. The experiment was set up in a completely randomized design (CRD) with four independent replicates.

Achievement/Findings:

The internode explants developed callus only. The highest percentage (75.00%) of response and the best growth of callus was observed in T₁₁ and the lowest percentage (17.19%) was found in control T₀. T₇ and T₈ and zeatin and IAA combinations produced better callus than the other treatments. Occasional shoot induction (1.56%) was observed in T₅. Tripathy (2014) found good callus from internode explants of grass pea. Shoot regeneration from calli induced from internode explants.
