Micropropagation of Dragon Fruit (hylocereus Undatus) (2017--2018)

Abstract/ Executive Summary:

Dragon fruit is being propagated by cutting mostly. But micropropagation may be a good means to produce more plantlets in a short time. For developing a micropropagation protocol of dragon fruit stem cuttings of BARI released dragon fruit were collected from the farm field of Pomology Division, HRC. Shoot tips and lateral portions of the newly developed stem were used as explant. Forty explants were cultured in shoot induction media. Among them, seven explants were responded to in culture media.

Objectives:

This study will help us to establish an efficient micropropagation protocol for dragon fruit so that more plantlets with the healthy shoot and root systems can be grown to fulfill the demand of increasing commercial production.

Methodology:

Cuttings of BARI Dragon Fruit-1 were collected from the Pomology Division of HRC. The cutting was planted in the farm field of the biotechnology division. New stem buds emerged from the planted cuttings. The young new stem of 10-15 days aged was detached from the plant and washed with tap water for 20 minutes then dip in ridomilgold @ 2g/l for 5 minutes again wash with tap water and another procedure was followed to disinfect the material. The young bud was sliced into 0.5-1.0 cm in thickness and cultured on MS (Murashige and Skoog, 1962) medium supplemented with BAP and NAA treated media. The pH of all media was adjusted to 5.8 before autoclaving. The inoculated cultures were incubated at 25±1⁰C under approximately 2000 lux by cool white fluorescent tubes for 16/8 h light/dark cycle in 60-70% relative humidity

Achievement/Findings:

The study was conducted to produce a plantlet of dragon fruit through micropropagation. Explants of the dragon fruit were cultured on the MS medium supplemented with different hormonal concentrations. Total of 40 explants were cultured. Among them, seven explants were responded and shoots were formed. This experiment will be continued for the next year to find the best media combination for prolific growth.
