Shoot tips of BARI Strawberry-2 and BARI Strawberry-3 were cultured on MS medium supplemented with different concentrations and combinations of BAP, Kn and GA3. Four different treatments T1, T2, T3, & T4, were tested for shoot multiplication and elongation. In case of BARI Strawberry-2, for shoot formation 1.0 mg/l BAP + 0.5 mg/l Kn +1.5 mg/l GA3 combination was found most suitable. Whereas in BARI Strawberry- 3, 0.5 mg/l BAP + 0.5 mg/l Kn +1.5 mg/l GA3 treatment was found most suitable for shoot formation. In rooting, ½MS medium without hormone produced maximum root than full MS medium. In field performance, BARI Strawberry-2 performed better in fruit number per plant while in single fruit weight, BARI Strawberry-3 performed better.

The present experiment is important to develop a suitable protocol for mass propagation of BARI strawberry varieties

BARI Strawberry-2 and BARI Strawberry-3 were collected from the fruits field of HRC, BARI and used as plant materials. Small shoot tip came out from runner were carefully removed by scalpel from Strawberry plants showing no virus symptoms. The explants were prepared by removing the outer layers of tissue from the shoot tips by the scalpel. The light greenish 2-3 cm shoot tips were surface sterilized with Ridomil Gold MZ 68WG for 5 min. to remove the fungal contamination. Then shoot tips were sterilized with Jet and Trix under running tap water for 5-6 min followed by 3-4 times washing with distilled water. After then shoot tips were sterilized with Clorox solution (60 ml Clorox: 40 ml distilled water) with 2 drops of Tween-20 for 3 min. followed by 3-4 times washing with sterile distilled water inside the laminar airflow cabinet. The dead tissue of sterilized explants was cut off to leave 1-2 cm portion containing an intact apex. The prepared explants were placed in MS media (Murashige and Skoog, 1962) with four concentrations (T1, T2, T3, and T4) with combinations of BAP, Kn and GA3. All cultures were incubated at 25 ± 10C with a 16 h photoperiod (2000 lux) provided by cool white fluorescent tubes. The pH of the medium was adjusted to 5.8.

Shoot tip explants of BARI Strawberry-2 and BARI Strawberry-3 were cultured on MS medium supplemented with four different concentrations and combinations of BAP, Kn and GA3. Shoot proliferation: Shoot initiation started within 6-7 days after inoculation. In BARI Strawberry-2, significant variation was observed in shoot number. The number of shoots per explants was higher in T1 and T4 treatment (11.00), although there was no significant difference between T1, T2 and T4 treatment. On the other hand, the lowest number of the shoot was obtained from T3 treatment (8.67). Shoot length was higher in T1 treatment (4.95 cm) and the lowest (3.20 cm) was obtained from T4 treatment. In leaf number, a significant difference was also observed. The maximum leaf (6.0) was found in T3 treatment and the lowest 12was obtained from T2 treatment (3.83). In BARI Strawberry-3, a significant difference was observed in all parameters. The number of shoots per explants was higher in T4 treatment (13.00). On the other hand, a lower number of the shoot was obtained from T1 treatment (4.50).  Shoot length was higher in T3 treatment (4.20 cm) and the lowest (3.52 cm) was obtained from T1 treatment.  The maximum leaf (5.17) was found in T4 treatment and the lower (4.17) was obtained from T2 treatment. Results indicated that the number of shoots was higher with an increase of BAP concentration in the medium. Because in this experiment only BAP concentration was increased, Kn and GA3 concentration were fixed. In vitro grown shoots were cultured in both MS and ½MS without hormone for rooting. Results showed that a hundred percent shoot produced roots. The shoots derived from T1 treatment cultured in ½MS medium produced maximum root (6.83) followed by T2 treatment in ½MS medium (6.58). On the other hand, a lower number of root (3.92) was observed from shoots of T4 treatment cultured in MS medium. The Highest root length (3.25 cm) was noticed from shoots derived from T3 treatment cultured in MS medium. The lowest root length (0.46) was observed from shoots of T4 treatment cultured in ½MS medium.  Results indicated that in strawberry rooting no growth hormone is needed. Well, rooted plantlets in the test tube were kept at room temperature for 2 to 3 days for hardening. After that, the plantlets were removed from the culture tubes and all the adhering media were washed carefully so that, the root damage was the least. The plantlets were then planted into small plastic pots containing sand, soil and decomposed cow dung at the ratio of 1:1:1 and covered with polythene bags to maintain high humidity for one or two weeks. After four weeks they were transferred to big plastic pots. The plantlets were successfully established in soil and ready to transplant in the field.

BARI Strawberry-2 and BARI Strawberry-3 tissue cultured plantlets were planted in the Biotechnology research field in October 2018. The field size was 755cm X180 cm. The planting method was bed planting. Line to line 60 cm and plant to plant 35 cm. A total of 120 plantlets were planted (each variety 60 plantlets).  All management practices and fertilizer dose were used according to the guideline of projukti handbook published by BARI.  Plants were growing well. Flowering and fruiting were also good.  Fruit harvest started on 9, January 2019 and finished on 29, April 2019. In BARI Strawberry-2, the maximum fruit number was 114, the minimum was 14 and the average fruit number per plant was 69. On the other hand, BARI Strawberry-3 produced a maximum of 67 fruit, a minimum of 4 fruit and the average fruit number per plant was 39 (Fig.1). In fruit weight, the Maximum single fruit weight was 31 g, minimum 5 g and average fruit weight was18.81 g in BARI Strawberry-2 while in BARI Strawberry-3, maximum single fruit weight was 39 g, minimum 6 g and average was 21.47 g. Results showed that in fruit numbers, BARI Strawberry-2 performed better while in fruit weight, BARI Strawberry-3 performed better.
