Molecular Characterization of Natural Edible Mushrooms in Bangladesh (2017-2018)

Abstract/ Executive Summary:

Ten natural edible mushrooms were collected from Khagrachari and Moulvibazar districts of Bangladesh during May-June, 2017. In the case of tissue culture, among 10 samples only one sample (i.e S-6) could be grown on PDA medium and was under domestication trials. It took about 19-22 days to fully colonize the substrate with mycelium. After the opening of spawn packets, it required 30-35 days to appear fruit bodies. The fruit bodies were harvested in two flushes and the calculated yield was 30 g and biological efficiency was 17%. In April 2018 four edible mushrooms were also collected from Rangamati district and their characteristics were done morphologically. Genomic DNA was extracted following standard procedures and preserved at -20⁰ C for further study.

Objectives:

The present investigation has been undertaken to identify and characterize the natural edible mushrooms of Bangladesh at the molecular level.

Methodology:

Collections of different wild edible mushrooms were done in April-June, 2017 and April 2018. The collection sites were forests, the local markets and nearby villages of Khagrachari, Moulvibazar and Rangamati districts of Bangladesh. The collected mushrooms were packed into butter paper bags and stored in the refrigerator until processing. Genomic DNA was extracted following standard procedures and preserved at -20º C for further study. During the year 2017, one sample (Sample 6)was purely cultured from the cap using the tissue culture method, maintained on Potato Agar slants and inoculated onto sawdust for spawn production. Spawn packets were prepared by sawdust mixed with wheat bran and Caco₃(0.2%). Water was added to make the moisture levels 65%. Polypropylene bags of 7"x10" sizes were filled with a 500 g mixture of the substrate. After autoclaving and cooling the bags were inoculated with the mother culture of sample 6 and incubated in the laboratory at about 28±2⁰C temperature. After completion of spawn running, the packets were cut into the top ad transferred to the culture house where a temperature of 25-30⁰C with 75-80% relative humidity. The moisture level was maintained by regular water spraying. Days for pinhead formation (DFPF), Days For the first harvest (DFFH), total yield and biological efficiency were calculated.
