Large-scale Multiplication of Planting Materials for BARI Released Banana Through Tissue Culture (2019--2020)

Abstract/ Executive Summary:

Tissue cultured banana plantlets have good demand among the farmers. Micropropagation is an effective method to produce large-scale banana plantlets within a short period of time. In this study, sword suckers of different banana varieties developed by BARI were collected from the farm field of Pomology Division, HRC and Hill Agriculture Research Station, Raikhali. Shoot tips were separated from those suckers and cultured on different shooting and rooting medium for in vitro production. Fifty suckers of different varieties were cultured. More than three hundred plantlets of different varieties were produced.

Objectives:

To produce large-scale banana plantlets through in vitro micropropagation.

Methodology:

Sword suckers of BARI Kala-3 and BARI Kala-4 were collected from the Pomology Division of HRC. Shoot tips were separated from those suckers and cultured on MS (Murashige and Skoog, 1962) medium supplemented with 5 mg/l BAP. The inoculated cultures were incubated at 25±1⁰C under approximately 2500-3000 lux by cool white fluorescent tubes for 16/8 h light/dark cycle in 60-70% relative humidity. Data on percent success of explant, bud initiation, no. of shoots /culture, length of shoots, days to root initiation, no. of root and length of roots/plant etc. were recorded.

Achievement/Findings:

The study was conducted to produce large-scale plantlets of different banana varieties through micropropagation. Shoot tip explants were cultured on MS medium supplemented with BAP 5 mg/l. Total 50 explants were cultured. Among them, 30 explants were survived and the rest were contaminated. Explant survival rate highly depends on the source of suckers. Epiphytic and endophytic organisms can cause severe losses to micropropagated plants at each stage of growth. From the 30 explants, different data were collected. In the case of BARI Kala -3, the explants were taken on an average of 58.2 days for multiple shooting. No of shoots per explant was 8.5. The average shoot length was 5.65 cm at the time of their sub-culture. On the other hand, BARI Kala-4 was taken on an average 56.7 days for multiple shooting. Number of shoots per explant was 7.3 and the average shoot length was 5.2 cm. Root formation data are presented in table-2. BARI kala-3 takes about 5 days and BARI kala-4 takes about 5.5 days. In BARI kala-3 and BARI kala-4 number of roots per plantlets was 4 and 5 respectively. In BARI Kala-3 and BARI Kala-4 length per plantlets were 5 and 6 respectively. More than three hundred plantlets were produced from BARI Kala-3 and BARI Kala-4 varieties. Due to pandemic Covid-19, plantlets production was hampered.
