In Vitro Regeneration of Soybean (2019--2019)

Abstract/ Executive Summary:

Leaf, Shoot tip, Cotyledon, Hypocotyl, Epicotyl explant of Soybean variety ‘Sohag’ were cultured on MS medium supplemented with different concentrations and combinations of hormones. Among the different explants, hypocotyl showed a better response for the formation of callus. Cotyledonary base explant produced direct shoot in 0.02 mg/l NAA+0.2 mg/l BAP and 2.0 mg/l BAP+0.5 mg/l NAA.

Objectives:

The present study has been undertaken to develop an in vitro regeneration protocol for soybean which will be used for future transformation work.

Methodology:

Seeds of soybean variety ‘Sohag’ were collected from Oilseed Research Centre, BARI, Gazipur. Soybean seeds were surface sterilized with 70% ethanol for 30 seconds, followed by three times with sterile distilled water. After then seeds were sterilized with Clorox solution (60 ml Clorox: 40 ml distilled water) and 2 drops of Tween-20 for 5-6 min followed by five times washing with sterile distilled water inside the laminar airflow cabinet. The sterilized seeds were placed in ½ MS media (Murashige and Skoog, 1962) incubated at 25 ± 1⁰C with a 16 h photoperiod (2000 lux) provided by cool white fluorescent tubes for about one week. Different seedling parts like a leaf, shoot tip, cotyledon, epicotyl, hypocotyl etc. were used as explants and placed in the MS media with different concentration of hormones

Achievement/Findings:

Different explants of soybean variety ‘Sohag’ were cultured on MS medium supplemented with different concentrations and combinations of hormones for three weeks. Among the different treatments, a hundred percent calli were formed from the explant hypocotyl with the treatment T_5,but the quality of callus from the treatment T₂showed better than that of treatment T_5.Different types of explants were tested under the treatments of T_6,T₇and T₈for callus formation. Whitish and friable callus was found from shoot tip, epicotyl base, epicotyl tip, and hypocotyl.The calli were transferred into the shooting media with different doses of BAP, NAA and GA_3,but no shoots formation was noticed. Direct shoot formation was found from the cotyledonary base under the treatments of T₆and T_7.
