Development of in Vitro Propagation Protocol for Gerbera (2019--2020)

Abstract/ Executive Summary:

An attempt has been made to develop a micropropagation protocol of gerbera. Capitulum, leaf, petal, sepal and flower stalk explant of pink, reddish, white and orange cultivars were cultured on MS medium supplemented with four different concentrations and combinations of BAP, NAA and IAA. Early primordial emergence (7 days) was observed in white cultivar on MS medium supplemented with 2.0 mg/l BAP + 0.5 mg/l NAA and 5.0 mg/l BAP + 1.0 mg/l NAA using capitulum explant. Direct shoot initiation was observed in 20 days from sepal explant both pink and white cultivar in 2.0 mg/l BAP + 0.5 mg/l NAA supplemented medium.

Objectives:

The present study has been undertaken to develop a suitable in vitro propagation of gerbera.

Methodology:

Capitulum, petal, leaf, sepal and flower stalk explants of pink, light pink, a reddish, white and orange cultivar of gerbera were collected from Bangabandhu Sheikh Mujibur Rahman Agricultural University (BSMRAU), Salna, Gazipur. The explants were surface sterilized under running tap water with Ridomil Gold 68 WG for 15 min. Thereafter, the explants were sterilized with jet and trix under running tap water for 15 min. followed by 3-4 times washing with distilled water. The explants were transferred to a sterilized container and brought to the laminar airflow cabinet and sterilized with clorox solution (60%) with 2-3 drops of Tween-20 for 10 min. followed by 3 to 4 times washing with sterile distilled water. The leaves were then prepared as cultured explant by slicing of leaves with/without midrib and cultured on medium in the lower part/upper part attached with the medium. Petal and sepal explants were prepared by pulling out directly using forceps one by one gently. Capitulum explants were cut into four parts with/without petals. Flower stalk explant also used in this experiment. In culture establishment, the sterilized prepared explants were inoculated in MS medium supplemented with different hormonal combinations of growth regulators BAP, NAA and IAA. All media were augmented with 30 g/l sucrose, 8 g/l agar and pH was adjusted in 5.8. Media were autoclaved in 121⁰C, and 15 psi for 20 min. All cultures were inoculated at 25 ± 1⁰C with a 16 h photoperiod (2000 lux) provided by cool white fluorescent tubes.

Capitulum, leaf, petal, sepal and flower stalk explant of different cultivars were cultured on MS medium supplemented with four different concentrations and combinations of BAP, NAA and IAA. Primordial emergence: The number of days taken for primordial emergence was minimum (7 days) with the treatment T₂ & T₃ in the white cultivar and T₂treatment in the orange cultivar using capitulum explant. This might be due to supplementation with cytokinin in the media which promotes shoot differentiation through protein and enzymatic activity. In cultivar reddish, the minimum days required for primordial emergence was 8d in T₁ treatment and the maximum was 12d in T₂ and T₃ treatment. Navya Swetha et al. (2017) observed in their study that 14.66 days required for primordial emergence. This might be due to varietal differences. Using flower stalk explant, the cultivar redish also showed primordial emergence in 11days in T₃ and 12 days in T₂treatment. Shoot initiation: For shoot formation, clumps of primordial were again subcultured in the same fresh medium. Shoot initiation was noticed from reddish and white cultivars respectively. On the other hand, direct shoot initiation was observed in 20d from sepal explant with pink cultivar in T₂ treatment and 23d in T₃ treatment. While in white cultivar, direct shoot initiation was noticed from T₂treatment using sepal explant in 20d. This variation might be due to variety and also explant type. Each plant and explant type has a specific response and different behavior in in vitro tissue culture
