Marker-assisted Transfer of Salt Tolerance (nax) Genes in Bangladeshi Wheat Varieties (2019--2020)

Abstract/ Executive Summary:

An experiment was conducted with a view to developing salt-tolerant wheat varieties using marker-assisted selection. Genotyping of some selected BC₃F₆progenies (basis on previous years field performance of BC₃F₅) of the cross of two salt-tolerant Australian wheat lines having Westonia background (Westonia 5907; Nax1 and Westonia 5924; Nax2) and two popular Bangladeshi wheat varieties BARI Gom-25 and BARI Gom-26 was done to confirm the presence of Nax genes. A total of 12 BC₃F₆ lines, 3 from each cross were selected comparing genotyping and field data. These 12 BC₃F₆ lines along with their parents were grown in a non-replicated trial for the increasing volume of seeds. Seeds of all lines were harvested and preserved for further trial and other research activities

Objectives:

The present experiment was initiated with a view to introgress the salt-tolerant Nax genes into selected Bangladeshi wheat varieties through marker-assisted backcrossing.

Methodology:

Thirty-six BC3F6 lines selected from the 72 BC₃F₅ progenies of 4 crosses among two Westonia lines (Westonia 5907; Nax1 and Westonia 5924; Nax2) obtained from CSIRO, Australia and two popular BARI varieties (BARI Gom-25 and BARI Gom-26) on the basis of field performance. Molecular genotyping of these thirty-six BC3F6 lines was done using Nax1 and Nax2 gene-specific PCR for confirmation of the genes in the selected lines. DNA was extracted from seedling leaves with the help of a plant DNA extraction kit. Selected 12 BC₃F₆ lines and their parents were grown in a non-replicated trial. Seeds were sown on 28-11-18. The plot size for each entry was 10’ X 6’ having 6 rows in each. Data were recorded on plant height (cm), the number of effective tillers per plant, days to 50% flowering, days to maturity, spike length (cm), 10 plant yield and plot yield (kg). Cultural management was carried out as required. BC₃F₇ seeds from each line were harvested manually and bulked separately.

Achievement/Findings:

In the PCR reaction of cross A (BARI Gom 25 X Nax1) it was observed that Bio-3, Bio-6, Bio-12 and Bio-15 showed specific bands for Nax1 region that means they were fixed whereas other genotypes show heterozygous bands. In the PCR reaction of cross C (BARI Gom 26X Nax1) it was observed that Bio-40, Bio-41, Bio-46, Bio-54 and Bio-54 showed a specific band for Nax1 region that means they were fixed whereas other genotypes show null and heterozygous bands. In PCR reaction of cross B (BARI Gom 25X Nax2) it was observed that Bio-20, Bio-22, Bio-23, Bio-26, Bio-27, Bio-31 and Bio-35 show specific bands for Nax2 region that means they were fixed whereas only Bio-30 was heterozygous bands. PCR reaction of cross d (BARI Gom 26X Nax2) it was observed that Bio-59, Bio-62, Bio-63, Bio-65, Bio-66, Bio-67 and Bio-68 show specific band with Nax2 region that means thy were fixed Seed germination of the selected 12 genotypes along with their parents was good and excellent plant establishment was achieved at the beginning of the experiment. However, unfavorable weather caused some damage to the crop. Recovery measures were undertaken and most of the crop was saved with satisfactory results. Complete data were collected from all 12 lines. Seeds of all 12 BC₃F₇lines were harvested and preserved for trials and further research.
