Conservation of Gerplasm in Field Gene Bank (2015--2016)

Abstract/ Executive Summary:

PGRC is maintaining 260 germplasm (213 accessions and 47 new collections) of 72 crops both indigenous and exotic germplasm in the field genebank. The fruit germplasm is mango litchi, banana, guava, jackfruit, jujube, aonla, bael, bilimbi, bullocks heart, etc. The vegetable germplasm are taro, yam, elephant foot, drumstick, etc. The exotic germplasm is rambhutan, pear, tamarind, coffee, passion fruit, dragon fruit, and gynura, etc. The intercultural practices were done as and when necessary. Some qualitative and quantitative traits of fig, carunda, sapota and ber were recorded for using as reference data. The field gene bank has been maintained since 1985 and continued for the following years.

Objectives:

There are two important methods of germplasm conservation or preservation namely, 1) in-situ conservation and 2) ex situ conservation. Field gene bank belongs to ex situ method where genetic variation is maintained away from its original location and samples of a species, subspecies, or variety are transferred and conserved as living collections.

Methodology:

The Field Gene Bank of Plant Genetic Resources Centre has been established with an aim to conserve vegetatively propagated and recalcitrant seeded plant germplasm. The plants were earthen up after rainy season. Fertilizer was applied in field gene bank crop in two installments in a year following fertilizer recommendation guide. Half of the installment was applied before and after rainy season.The plants were watered and weeded as and when necessary. There was no pest and disease infestation during the period. Yam germplasm were harvested and replanted for maintaining after every two years. The other germplasm were maintained accordingly. The corm of Ol kachu was also harvested and replanted for maintaining every year. There was only one established fig germplasm in PGRC Field Genebank. The doses of fertilizer were 30 kg cowdung, 1kg urea, 500gTSP, 500g MP and 300 g zypsum. The fertilizer was applied in two installment, half of the installment before and after rainy season. The age of the fig tree was more than 20 years. The data recorded Sept. 2015 to May 2016 during 2015-16. There was a twenty years old (approx.) established germplasm of carunda at PGRC field Genebank. Recommended fertilizer dose was applied in two installments. One installment applied before rainy season and other after rainy season. Other cultural practices were done as when necessary. The data was recorded from July to May during 2015-16. There were one sapota and three ber germplasm in PGRC Field Genebank. Data were recorded as per descriptor published by NBPGR (Mahajon et. al., 2002).

A total of 260 (213 accessions and 47 germplasm) of indigenous and exotic are conserved as a field gene bank. The accessions and germplasm are maintained as living plants, either in the field or in pots in the nursery. PGRC has developed five field gene bank for maintaining germplasm in PGRC, Gazipur; Agricultural Research Station, Burirhat, Rangpur, Hill Research Centre, Khagrachari, Regional Research Station, Rahmatpur-Barisal and Citrus Research Station, Jaintapur, Shylhet. A total of 260 (213 accessions and 47 germplasm) of 72 crops is maintaining at the field genebank. The field gene bank acts as a reliable source of indigenous and exotic germplasm for researchers, plant breeders, NGOs, and farmers for crop improvement.
