Conservation of Germplasm in Active and Base Collection (2016--2017)

Abstract/ Executive Summary:

Plant Genetic Resources Centre (PGRC) acts as a repository for the BARI mandate crops namely cereals (except rice), pulse, oilseed, vegetables, fruits, spices, tuber crops, etc. Six hundred and fifty-five germplasm of newly collected, 642 newly assigned accession and 478 existing accessions from regeneration and characterization were conserved in genebank during 2016-17. The accessions were conserved in medium-term storage at + 4 to + 6⁰C and long-term storage at -18 to -22⁰C. Generally, the seeds were dried at 6-8% moisture content before storing. The centre has been maintained 10,799 accessions of 84 different agro-horticultural crops in the gene bank. Among them, 1768 accessions were cereals, 3529 pulses, 552 oilseeds, 4205 vegetables, 404 spices, 283 fruit, and 58 other crops. Viability, quantity, and moisture content were checked before the conservation of germplasm. Accessions less than 85% viability and/or less quantity of seeds were regenerated.

Objectives:

To overcome the problem the seeds of landraces, cultivars, and their wild relatives are collected, characterized, regenerated, and conserved for their future utilization.

Methodology:

Six hundred and fifty-five germplasm from different districts were registered according to the collection trip during 2016-17. The collected germplasm was cleaned and dried. A registration number was assigned to each germplasm and was kept in active collection at 4⁰C. Six hundred and forty-two germplasm were assigned new accessions from the germplasm that were characterized and or regenerated during 2015-16 and 2016-17. 'BD' code was used in the accession number. In addition, 478 accessions have conserved those accessions which were characterized and or regenerated during 2015-16. After regeneration and accessioning, the original samples were maintained in base collection for future use and regeneration. The viability, quantity and moisture content of each accession were checked before conservation in the genebank. Viability, quantity, and moisture content of all characterized and or regenerated germplasm from 2015-16 and 2016-17 were checked. All the germplasm were conserved in the genebank. The accessions having less than 85% and or less seed weight germplasm will be regenerated in the following year. In 2016-17, six hundred forty-two (642) germplasm were assigned new accessions. Among them, 16 cereals, 15 pulses, 76 oil seeds, 211 vegetables, 137 spices, and 187 fruit crops. Six hundred and fifty-five germplasm from different districts were registered and was kept in active collection at 4⁰C (Table 2). Up to June 2017, a total of 10799 accessions of 84 different agri-horticultural crops were maintained as the active collection and base collection.
