Monitoring of Germplasm in Active and Base Collection (2016--2017)

Abstract/ Executive Summary:

Germination percent of 488 accessions among 22 crops were tested from active collection temperature at 4-6⁰C for those conserved for 8 years. The viability test was conducted on germination paper for 8 to 21 days during 2016-17. The crops were cereals (wheat, proso millet and teff), pulse (grasspea, mung bean, chickea pea, black gram and field pea), oilseed (linseed), vegetable (grain amaranth, bitter gourd, red amaranth, snake gourd, okra, bottle gourd and pumpkin), spices (sulfa, fenugreek, black cumin and coriander) and jute. Out of 488 accessions, below 40% germination was found in 47 accessions, 40 to 80% germination in 118 accessions and 81 to100% germination in 323 accessions. Similarly, germination percent of 288 accessions among hyacinth bean, pumpkin, ash gourd and bottle gourd were tested from base collection temperature at -18 to -22⁰C those conserved for 20 years. Out of 288 accessions, below 40% germination was exhibited in 59 accessions, 40 to 80% germination in 81 accessions and 81-100% germination in 148 accessions. Seed quantity of 1500 accessions (BD-1 to BD-1600) of wheat, maize, foxtail millet, prosomillet, teff, hyacinth bean, pumpkin, bitter gourd, bottle gourd and ash gourd from active and base collection were monitored. The accessions having less than 80% germination and or less quantity will be regenerated in the following year.

Objectives:

The objective of monitoring is to determine whether regeneration or multiplication of accession is required.

Methodology:

The accessions of different crops such as cereals (wheat, proso millet, and teff), pulse (grasspea, mung bean, chickpea, pea, black gram, and field pea), oilseed (linseed), vegetable (grain amaranth, suji, bitter gourd, red amaranth, snake gourd, okra, bottle gourd, and pumpkin), spices (sulfa, fenugreek, black cumin, and coriander) and jute were tested for germination from the active collection of 2008-09 Similarly, germination percent of 288 accessions among hyacinth bean, pumpkin, ash gourd, and bottle gourd were tested from the base collection (at -18 to -22⁰C) those conserved during 1996-97. From July 2016 to June 2017, all the samples were put into germination paper for viability testing. The number of normal seedlings, abnormal seedlings, dormant and dead seeds was counted and viability (%) was calculated. Seed quantity of 1500 accessions (BD-1 to BD-1600) of wheat, maize, foxtail millet, prosomillet, teff, hyacinth bean, pumpkin, bitter gourd, bottle gourd, and ash gourd from the active and base collection were monitored.

Achievement/Findings:

Out of 488 accessions, 47 accessions were in <40% germination, 118 accessions between 40-80%, and 323 accessions between 81-100% germination. About 67% of accessions were showed viability between 81-100% after 8 years of conservation of the germplasm from the active collection. Similarly, germination percent of 288 accessions among hyacinth bean, pumpkin, ash gourd, and bottle gourd were tested from the base collection (at -18 to -22⁰C) those conserved during 1996-97. Out of 288 accessions, 59 accessions were showed below 40% germination, 81 accessions showed between 40 to 80%, and 148 accessions in 81-100% germination. About 52% of accessions were showed viability between 81 to 100% after 20 years of conservation of the germplasm in base collection. Seed quantity of 1500 accessions (BD-1 to BD-1600) of wheat, maize, foxtail millet, prosomillet, teff, hyacinth bean, pumpkin, bitter gourd, bottle gourd, and ash gourd from the active and base collection was checked.
