Conservation of Germplasm in Active and Base Collection (2017--2018)

Abstract/ Executive Summary:

Plant Genetic Resources Centre act as a national repository for the BARI mandate crops namely cereals (except rice), pulse, oilseed, vegetables, fruits, spices, tuber crops etc. Three hundred and fifty-two germplasm of newly collected and 1246 germplasm from regeneration and characterization were conserved in genebank during 2017-18. The accessions were conserved in active collection at + 4 to + 6⁰C and base collection at -18 to -22⁰C. Generally, the seeds were dried at 6-8% moisture content before conservation. The centre has been maintained 10,799 accessions of 84 different agro-horticultural crops in the gene bank. Among them, 1768 accessions were cereals, 3529 pulses, 552 oilseeds, 4205 vegetables, 404 spices, 283 fruit, and 58 other crops. Variability, viability, quantity, and moisture content were checked before the conservation of accessions. Accessions less than 85% viability and/or less quantity of seeds were regenerated.

Objectives:

The center has been collected and maintained a number of different agro-horticultural crops in the gene bank and field gene bank since 1987. The objective of the study is to retain the viability of germplasm for a longer period and future use.

Methodology:

One thousand two hundred and forty-six germplasm of 29 crops were conserved in the active and base collection unit of PGRC during 2017-18. The seeds were produced from characterization and regeneration during 2016-17 and 2015-16. 'BD' code is used in the accession number. Variability, viability, quantity, and moisture content of accessions were checked before conservation in the genebank. Laminated aluminum foil packets and polyethylene bags were used in base and active collection, respectively. The storage temperature was maintained at 4 to 6⁰C and -18 to -22⁰C for active and base collection, respectively. Three hundred and fifty-two germplasm from different districts were registered according to the collection trip during 2017-18. The collected germplasm was cleaned and dried. A registration number was assigned to each germplasm and was kept in active collection at 4⁰C. Among them, 999 and 961 accessions were conserved in active and base collection, respectively. Variability, viability, quantity, and moisture content of accessions were checked. The accessions having less than 85% and or less seed weight germplasm will be regenerated in the following year. Three hundred and fifty-two germplasm from different districts were registered and were kept in active collection at 4⁰C. Some new collections like barnyard millet, nati shak, tal makhna, lubna, subrang and bui kumra were conserved in active collection. Up to June 2018, a total of 10799 accessions of 84 different agri-horticultural crops were maintained as the active collection and base collection. Among them, 1768 accessions are cereals, 3529 pulses, 552 oilseeds, 4205 vegetables, 404 spices, 283 fruits, and 58 other crops
