Monitoring of Germplasm in Active and Base Collection (2017--2018)

Abstract/ Executive Summary:

Germination percent of 696 accessions among 12 crops were tested from active collection temperature at 4-6⁰C those conserved for 3 to 20 years. The viability test was conducted on germination paper during 2017-18. The crops were wheat, sorghum, prosomillet, oat, pigeon pea, sesame, soybean, linseed, black cumin, hyacinth bean, rosel, and tomato. Among the 696 accessions, 61% , 12%, 9% and 18% accessions showed 85 to 100%, 70 to 84%, 40 to 69% and below 40% viability, respectively. Germination percent of 589 accessions among wheat, foxtail millet, prosomillet, maize, sorghum, pigeon pea, bitter gourd, soybean, and sweet gourd were tested from base collection temperature at -18 to -22⁰C those conserved for 6 to 28 years. Out of 589 accessions, below 40% germination were exhibited in 27% accessions, 40 to 69% germination in 6%accessions, 70 to 84% germination in 10% accessions and 85 to 100% germination in 57% accessions. Seed quantity of the accessions was monitored. The accessions having less than 70% germination and or less quantity will be regenerated in the following year.

Objectives:

The objective of monitoring is to check the viability and quantity of conserved germplasm whether regeneration or multiplication of accession is required.

Methodology:

The accessions of 12 crops such as wheat, sorghum, prosomillet, oat, pigeon pea, sesame, soybean, linseed, black cumin, hyacinth bean, rosel and tomato were tested for germination from active collection from 1997 to 2016. Similarly, germination percent of 589 accessions among the wheat, foxtail millet, proso millet, maize, sorghum, pigeon pea, bitter gourd, soybean, and sweet gourd were tested from base collection temperature at -18 to -22⁰C those conserved during 1989 to 2012. From July 2017 to June 2018, all the samples were put into germination paper for viability testing. The number of normal seedlings, abnormal seedlings, dormant and dead seeds was counted and viability (%) was calculated. Seed quantity of accessions from the active and base collection was monitored.

Similarly, germination percent of 589 accessions among wheat, foxtail millet, proso millet, maize, sorghum, pigeon pea, bitter gourd, soybean, and sweet gourd were tested from base collection temperature at -18 to -22⁰C those conserved during 1989 to 2012. Out of 589 accessions, 159 accessions were in <40% germination, 37 accessions between 40-69%, 59 accessions between 70-84% and 334 accessions between 85-100% germination.
