In Vitro Conservation of Potato (2017--2018)

Abstract/ Executive Summary:

The experiment was conducted with 8 varieties of potato namely Diamant, Cardinal, Dheera, Lady Rosseta, Quency, Almina, BARI Alu-33, BARI Alu-37, and BARI Alu-41 at the In vitro Conservation Laboratory of PGRC, BARI, Joydebpur, Gazipur during 2017-18. It is a medium-term study of in vitro conservation on potato varieties which had been initiated in 2014. The experiment was conducted at 24⁰C to evaluate the performance of in vitro conservation. Nodal explants of potato varieties were cultured for 5 months on MS medium containing 2% sorbitol. The varieties initiated roots within 15 days and shoots within 16 days. The varieties produced 28 to 38 leaves, 5 to 12 shoots, and 5 to 19 roots at six-month culture. The maximum shoot length (13 cm) was exhibited in Lady Rosseta and the minimum shoot length (8.1 cm) was obtained from BARI Alu-37.

Objectives:

They need in vitro conservation. Therefore, the objective of this study is to conserve the potato varieties for in vitro conditions.

Methodology:

The experiment was conducted with 8 varieties of potato such as Diamant, Cardinal, Dheera, Lady Rosseta, Quency, Almina, BARI Alu-37, and BARI Alu-41 at the In vitro Conservation Laboratory of PGRC, BARI, Joydebpur, Gazipur during 2017-18. This is a medium-term study of in vitro conservation on 8 potato (Solanum tuberosum L.) varieties that had been initiated during 2014. The varieties have been maintaining at the laboratory for four years. Plantlets were developed in MS medium and approximately 5 weeks aged plantlets were used in this experiment. MS media containing 2% sorbitol was prepared. The pH of the medium was adjusted to 5.8 before autoclaving and the media were solidified with agar @ 8 g/l. The medium was autoclaved at 1.06 kg/cm² pressure and 121°C for 20 min. Nodal explants were prepared under Laminar Air Flow aseptically and inoculated one explant into a long test tube (25 x 150 mm) containing 15 ml MS medium. Each nodal explant having one node and one leaf pair was cultured. The explants were incubated for five months in a culture room at 24 ± 1°C under about 500 lux light provided by cool-white fluorescent tubes maintaining a 16/8-hr alternate light/dark cycle and 60-70% relative humidity. The experimental setup was in a completely randomized design with five replications and each replication consisted of 5 explants. Mean data were calculated.

Achievement/Findings:

The varieties produced 85 to 100% roots within 8 to15 days and 80 to 100% shoots within 9 to 17 days. After five months conserved in MS medium containing 2% sorbitol, BARI Alu-28 produced the maximum leaflets (38) while the BARI Alu-41 produced the minimum leaflets (28). The varieties produced 5 to 12 shoots and 5 to 19 roots. The maximum shoot length (13 cm) was exhibited in BARI Alu-28, and the minimum (8.1 cm) was obtained from BARI Alu-37. Among the varieties, BARI Alu-41 performed minimum shoot proliferation through node culture. Most of the varieties produced micro tuber at 5 months cultured on the medium. The present study demonstrates that MS medium with 2% sorbitol showed good performance in vitro conservation of potato varieties. The minimal shoot proliferation was achieved in BARI Alu-37 through node culture
