In Vitro Conservation of Mint (2017--2018)

Abstract/ Executive Summary:

The experiment was conducted with three genotypes viz. MP-1, MP-2, and MP-3 of mint (Mentha spp.) at the In vitro Conservation Laboratory of Plant Genetic Resources Centre, BARI, Joydebpur Gazipur. It is a medium-term study on in vitro conservation at 24 ± 1°C on 3 mint genotypes which had been initiated during 2014. Nodal explants of three mint genotypes were cultured on MS medium containing 2% sorbitol for five months. The maximum leaflets (86), shoots (9), shoot length (12.34 cm), roots (13), and root length (3.5 cm) were observed in MP-2. The minimum number of leaflets (58), shoot, and roots (4) was found in MP-1. While the MP-3 produced the minimum shoot length (10.5 3cm) and root length (3.0 cm). MP-2 performed better in the growth of in vitro conservation.

Objectives:

Therefore, an attempt has been made to increase the in vitro conservation period of mint genotypes.

Methodology:

The experiment was conducted with three genotypes viz. MP-1, MP-2, and MP-3 of mint at the In vitro Conservation Laboratory of Plant Genetic Resources Centre, BARI, Joydebpur Gazipur. The genotypes have been maintaining at the Laboratory for four years. Basal media contained MS macro and microelements including vitamins (Murashige and Skoog 1962). Four weeks aged plantlets were taken on sterilized petridish. Nodal segments with two leaflets were cut aseptically under laminar flow and these segments were used as explants. MS basal salts 44.0519 mg/l, 30 g sucrose, and 20 g sorbitol were dissolved in distilled water and mixed well. The pH of the medium was adjusted to 5.8 before autoclaving and the media were solidified with agar @ 8 g/l. Fifteen ml of media was poured into each culture tube (25 x 150 mm). The medium was autoclaved at 1.06 kg/cm² pressure and 121°C for 20 min. One nodal explant was placed vertically on the medium keeping the position of the tip upsides and gently pressed into the surface of the sterilized culture medium. The nodal explants were incubated for three months in a culture room at 24 ± 1°C under about 2000 lux light provided by cool-white fluorescent tubes maintaining a 16/8 hr alternate light/dark cycle and 60-70% relative humidity. The culture tubes were checked daily to note the morphogenic development and the data were recorded on 5 parameters. The experimental setup was in a completely randomized design with five replications and each replication consisted of 5 explants.

Achievement/Findings:

After five months of culture in MS containing 2% sorbitol, the maximum number of leaflets (86), shoots (9), shoot length (12.34 cm), roots (13), and root length (3.5 cm) was observed in MP-2. The varieties produced roots within 11 to 13 days and shoots within 9 to 11 days. The minimum number of leaflets (58), shoots, and roots (4) was found in MP-1. While the MP-3 produced the minimum shoot length (10.5 cm) and root length (3.0 cm). MP-2 performed better in the growth of in vitro conservation. MP-2 performed better than the remaining genotypes for growth in mint through node culture. MS medium containing 2% sorbitol could be used in vitro conservation of mint
