Nodal explants of five yam accessions (Dioscorea  alata) were cultured for six months at 22±10C on MS media to access the influence of two concentrations of Kinetin (Kn) with Indol Acetic Acid (IAA) on in vitro conservation. Five and 10 mg/l concentrations of Kn along with 2 mg/l  IAA and 3% sucrose were used. The accessions were produced shoots and roots within three weeks. The maximum number of leaves (15.7) was obtained from 10 mg/l Kn and the minimum (13.6) from 5 mg/l kn. The maximum shoot length (6.5 cm) was found in 10 mg/l Kn and minimum (6.1cm) in 5mg/l Kn.  The accessions produced 1.6 and 1.9 shoots, 12.3 and 13.1 roots in 5mg/l Kn and 10 mg/l Kn, respectively. Maximum root length (6.7 cm) was obtained from 10 mg/l Kn. The accession, BD-7848 performed better for shoot proliferation through node culture.

The objective of this study is to develop the in vitro regeneration protocol of yam accessions.

The work was carried out at the In vitro Conservation Laboratory of Plant Genetic Resources Centre, BARI, Joydebpur Gazipur.  The experiment was conducted with 5 accessions of yam. The accessions were obtained from different districts in Bangladesh. Tubers of all accessions were grown in the field of PGRC. The vines of the accessions were grown enough long. Nodal explants of about 3 cm length were used after removal of the leaf and trimming of the petiole. They were disinfected with 0.3% Bavistin for 10 minutes and then washed with sterilized distilled water. Then it was surface disinfected with 70% ethanol and then with sodium hypochlorite (effective chlorine concentration 3 %) and Tween 20 (2-3 drops) for 15 min, then rinsed three times in sterile distilled water. Basal media contained MS macro and microelements (Murashige and Skoog 1962) and 0.1 mg/l thiamine. In order to assess the capacity of the cultures to produce shoot and roots, the basal MS medium was supplemented with 5 mg and 10 mg/l Kn along with 2 mg/l IAA and 3% sucrose. The pH was adjusted to 5.8. The medium was autoclaved at 121 °C for 20 min. Thirty ml medium was taken into the glass bottle. Nodal explants were excised aseptically and inoculated into a glass bottle covered with aluminum foil and a plastic cap.  One explant was cultured in each glass bottle. The explants were cultured at 22 °C cells for six months under the fluorescent light of 35-55 µmol/m2 /s using 16-h illumination. The experimental setup was in a completely randomized design with five replications and each replication consisted of 5 explants.

The accessions produced shoots and roots within three weeks. After six months cultured in MS medium containing 10 mg/l Kn, BD-7848 produced the maximum leaves (23.7) while BD-7864 produced the minimum leaves (11.7).  Considering five accessions, the maximum number of leaves (15.7) were obtained from 10 mg/l Kn and the minimum (13.6) from 5 mg/l kn. However, the maximum shoot length (6.5) was found in 10 mg/l Kn and minimum (6.1) in 5mg/l Kn.  The accessions produced 1.6 to 1.9 shoots after six months of culture. The accessions produced 4.8 to 7.8 cm roots in 5mg/l Kn and 5.6 to 7.8 cm roots in 10mg/l kn Among the accessions, BD-7848 performed better for shoot proliferation through node culture. A lower concentration of Kn along with IAA and sucrose was the better treatment for leaf and root development in yam accessions. This technique could be used for in vitro conservation of yam accessions.
