Conservation of Germplasm in Active and Base Collection (2019--2020)

Abstract/ Executive Summary:

Plant Genetic Resources Centre (PGRC) acts as a germplasm storehouse of the BARI mandated crops viz. cereals, pulses, oilseeds, vegetables, fruits, spices, and other crops etc., since 1987. The accessions were conserved in medium-term storage (at 4 to 6⁰C) and long-term storage (at -18 to -22⁰C). Viability (germination %), quantity, and moisture were checked before conservation. Accessions having less than 80% viability and/or less quantity of seeds were regenerated. Generally, the seeds were dried at 6-8% moisture content before storing. Till now, the Centre has conserved 11188 (eleven thousand one hundred and eighty-eight) accessions of 82 different crops in its gene bank. Among them, 1768 accessions were cereals, 3553 pulses, 602 oilseeds, 439 spices, 4486 vegetables, 283 fruits, and 57 other crops. In 2019-20, a total of 107 germplasm was assigned as new accession and were conserved in the gene bank of PGRC.

Objectives:

To overcome the problem the seeds of landraces, cultivars and their wild relatives are collected, characterized, regenerated, and conserved for their future utilization.

Methodology:

The conservation of germplasm in their natural habitat is called in situ and away from their natural habitat is ex situ conservation. PGRC conserved PGR via ex situ conservation techniques including seed gene bank and field gene bank (Bioversity International, 2007).

PGR Collection: A team of PGRC scientists collected germplasm every year from different districts and were registered according to the collection trip year. The collected germplasm was cleaned and dried. A registration number was given to each germplasm and was kept in active collection at 4⁰C.

The procedure of assigning BD accession: Normally, the seed sample size of 7,000 for self-pollinated and 17,000 for cross-pollinated crops along with 80% and above viability were the major prerequisite for getting an accession number. 'BD' for Bangladesh code is used before the accession number.

Conservation of base collection: The base collection is a long-term storage facility. The initially collected seed accession sample is used as a benchmark for monitoring changes in the genetic identity and diversity of the accession, over time and after cycles of regeneration, that may arise due to genetic drift. The facility should ensure that an accession’s seed viability remains at acceptable levels for at least 50 years, or at least one cycle (Furman et al., 2009) of the regeneration of the accession.

Conservation of active collection: The active collection consists of accessions that are designated as available for distribution and hence are accessed more frequently than those in the base collection. The active collection is intended to preserve germplasm viability in medium-term storage conditions for a maximum of 20 years or for half of one regeneration cycle (Furman et al., 2009) in the base collection

Plant Genetic Resources Centre (PGRC) has conserved 11188 (eleven thousand one hundred and eighty-eight) accessions of 82 different BARI-mandated crops in its gene bank. Among them, 1768 accessions were cereals, 3553 pulses, 602 oilseeds, 439 spices, 4486 vegetables, 283 fruits, and 57 other crops. A total of 107 germplasm were newly added to its previous conservation. Among them, vegetables (28) spices (7) oilseeds (50), and pulses (22) were included. These 107 germplasms were assigned as new accession with BD code and were preserved in the seed gene bank of PGRC (active and base collection). Germplasm having germination capacity of more than 80%, sufficient well-developed cleaned seed, and <10% moisture content for getting new accession. All the germplasm was conserved in the gene bank. The accessions having less than 80% and or less seed weight germplasm will be regenerated in the following year.
