The experiment was done to investigate seed reserved utilization and seedling growth in two wheat genotypes (BARI gom 26 and Pavon 76) under normal (25°C) and high (35°C) temperature condition. Higher seed reserve utilization efficiency (SRUE) was recorded in BARI gom 26 (1.05 mg/mg) and lower in Pavon 76 (0.98 mg/mg) due to high temperature stress. Under high-temperature stress condition, the contribution of reserved seed materials continued for a relatively longer period in BARI gom 26 than Pavon 76. Daily dry matter accumulation in seedling (shoot + root) was higher in BARI gom 26 compared to Pavon 76 during 7-14 day duration of autotrophic seedling growth phase compared to that found at 25°C temperature. In terms of seed reserve utilization efficiency, BARI gom 26 was found to be superior variety for growing under warmer environment.

In a preliminary study, Hasan et al.  (2004) and Cargnin et al. (2006) found that heat tolerant wheat genotypes exhibited lesser loss of seed dry matter than sensitive genotypes during germination. The present study was therefore, conducted to determine the seed endosperm use efficiency and seedling growth of wheat genotypes in response of elevated temperature and that heat tolerance may be associated with the heat tolerance in the autotrophic plant after germination.

The experiment was conducted at the Crop Botany Laboratory, Bangabandhu Sheikh Mujibur Rahman Agricultural University (BSMRAU), Gazipur during the year 2012. Experiment was conducted by using two factor completely randomized design. Two wheat genotypes namely BARI gom 26 and Pavon 76 were collected from Wheat Research Centre (WRC) of BARI, Joydebpur and Wheat Research Centre (WRC) of BARI, Dinajpur were used for the present study and considered as factor A. Two temperature regimes for growing condition (35°C/25°C and 25°C/15°C day-night temperature) were considered as factor B.

Determination of seed reserve utilization efficiency

Before placement of seed for germination, the seeds of a genotype were thoroughly mixed and moisture percentage was determined gravimetrically using a portion of the seeds, the remaining seeds were used for the experiment. Individual weight of 25 seeds for each genotype was taken and was placed sequentially according to the marking on plastic tray filled with pure sand with adequate moisture at a time. Then the trays were kept in seed germinator (ATTEMPTER, Advantec, Japan) at 25°C and 35°C respectively. For each temperature four batches of trays each containing 25 seeds was used. Water was added to the trays when necessary. After five days of sowing, Hoagland's nutrient solution was applied rather than water. At 3rd day after placement for germination, 25 seedlings were sampled. Then shoot, root and remaining seeds were dried separately at 70°C for 72 h and weight were recorded by using analytical balance (AND Electronic Balance Model ER 180A A & D Company Limited, Tokyo, Japan). Seed reserve utilization efficiency in the present study is defined as the amount of shoot and root dry matter (mg) produced from 1 unit (mg) of dry seed weight that was loss as respiration. Thus higher the value of seed reserve utilization efficiency (SRUE), the higher is the efficiency of seed as more seed reserves would be used for producing roots and shoots.

Amount of seed material loss as respiration (SMR) was calculated as Hasan et al (2004) by the following formula-

SMR = SDW- (SHW +RTW +RSW) 

Where,

SDW = Seed dry weight before germination SHW = Shoot dry weight

RTW = Root dry weight

RSW = Remaining seed dry weight

Total dry weight (TDW) gained everyday was calculated as-
                         TDW= SHW + RTW+ RSW

           Statistical analysis

The data were analyzed by two factor completely randomized design using computer operated MSTAT-C program. The treatment means were compared using Duncun's New Multiple Range Test (DMRT) at 5% level of significance and simple data were calculated by using Microsoft Excel 2001.

During germination BARI gom 26 maintained higher seed reserve utilization efficiency, producing 1.05 mg of seedling dry matter at the cost of 1 mg of reserve seed materials under higher germination temperature in comparison with Pavon 76 that produced 0.98 mg seedling dry matter/mg seed material respired. The heat tolerance in terms of seed reserve utilization efficiency during germination might be used as selection criteria for screening wheat genotypes against heat stress in laboratory.
