In Vitro Propagation of Mulberry (morus Alba L.) : Effects of Cytokinin Types and Tyrosine

Abstract/ Executive Summary:

In vitro clonal propagation of Moms alba cv S1 was achieved by culturing nodal explants and shoot apices of mature trees on Murashige and Skoog's medium supplemented with different cytokinins alone or incombination with tyrosine. Maximum frequency of explants produced shoots and highest number of shoots per explant were obtained from cultures grown on media supplemented with BAP. Further increase in shoot number was obtained by addition of tyrosine. Yield from nodal explants was greater than from shoot apices. IBA was found to be superior for root induction. Rooted plantlets were acclimated and successfully established in soil.

Objectives:

To the effect of different types of cytokinins on multipule shoot proliferation in mulberry (morus alba L.) effects of cytokinin was also studied since there is no report in this regard.

Methodology:

Shoot apex and axillary buds were taken from 10-12 year old tree. The explains were kept under running tap water for 1 h to washout the fungal and bacterial spores adhere on its surface. Next, the explants were dipped in Tween 80 BDH solution for 20-30 min and than washed thoroughly with distilled water. The explants were then surface sterilized with HgCl₂ (0.1% w/v) and cultured on MS (Murashige and Skoog, 1962) medium supplemented with 6-benzylamino purine (BAP), Kinetin (Kn), 2-isopentenyladenine (2iP) and zeatin (Zn) each 2.0 mg/l with or without 50 mg/l lyrosine. The explants were subcultured in fresh medium in every 4 weeks and data were recorded after 4 weeks of second subculture. The pH of the medium was adjusted to 5.8-6.0 before jellying the medium with 0.7% Difco-bacto agar and autoclaved at 121°C for 15 min at 15 Ibs pressure/cm2. All cultures were maintained in a culture room at 27.2°C with a relative humidity at 50-60 percent and were exposed to white Philips fluorescent tube light for 16 hours per day. After 6 weeks of culture the shoots that reached upto 2-3 cm long were excised and cultured on half strength MS media using three different auxins namely, indolebutyric acid (IBA), indoleacitic acid (IAA) and naphthaleneacitic acid (NAA), each 1.0 mg/1. Data on different rooting parameters were recorded after 5 weeks of culture in rooting media.

Achievement/Findings:

This research work highlights the potentialities of in vitro shoot proliferation from shoot apex and axillary bud of mature tree in mulberry. BAP was found to be most suitable cytoknin for multiple shoot proliferation could be accelerated by adding tyrosine in the media. The results correspond with the work of rahman et. al. (1985) and Hossain et. al. (1990). Yield from nodal explants was greater than from shoot apices.
