Effect of Different Cytokinins on in Vitro Propagation of Mulberry (m. Sp.) Through Meristem Culture.

Abstract/ Executive Summary:

Meristems from BM3 variety of mulberry (Morus sp.) were cultured for shoot proliferation and root proliferation and root induction in MS semisolid medium supplemented with different types and concentrations of cytokinins. Excised merstems were cultured onto MSO and | MSO (without growth regulator) liquid medium for 3 weeks. After 3 weeks, established meristems were transferred from liquid MSO and -j MSO medium to semisolid MS medium with different concentrations of cytokinins for shoot proliferation. Among the different treatments, MS+1.5 mg/l BAP showed good response of shoot proliferation. The reproducible protocol can be used for production of virus free mulberry plants.

Objectives:

To established an in virto propagation of Morus.sp. (BM3) through meristem culture.

Methodology:

Apical shoot tips (1-3 cm) of BM3 variety of mulberry (8-10 years old low-cut tree) were excised with the help of sharp blade and collected in bottle containing distilled water with few drops of savlon and then taken to the laboratory. The explants were washed thoroughly under running tap water for Ih to washout the fungal and bacterial spores adhered on its surface then explants were dipped in tween (BDH) solution for 20-30 min. with constant shaking. Next, the explants were washed with gradual change of distilled water. The explants were then surface sterilized with HgCl₂ (0.1% w/v) by gentle shaking for 5-8 min. and shoot tips were then washed 5-7 times with sterile D/W. In front of a running laminar air flow cabinet, shoot tip was hold in one hand under the stereo microscope with help of a pair of forceps and the immature leaves and leaf primordia was snapped off with slight pressure with the help of a needle. The meristems were (0.1-0.25) mm) excised and immediately carefully placed on the filter paper bridge of the culture tubes containing liquid growth regulator free MS and MSO medium. The established isolated meristems were sub-cultured for shoot proliferation on semi solid MS (Murashing & Skoog 1962) medium supplemented with different concentration of BAP and KIN for 5 weeks and data were recorded after 5 weeks second sub-culture. The PH of the medium was adjust to 5.7±0.1 before adding the agar (0.7% Difo-bacto) and autoclave at 121°C for 15 min at 15 abs. pressure/cm2. Finally the culture were maintained at 28±1°C with a 16h photoperiod.

Achievement/Findings:

The result of the present investigation highlight the potentialities of in vitro shoot proliferation from meristems of low-cut type of tree in mulberry. BAP was found to be most suitable growth regulator for shoot proliferation. The result corresponds with the work of Islam et al. (1993), Rahman et al. (1985) and Hossainetal.(1990).
