In Vitro Shoot Proliferation of Morus Alba Cv. Bm2 : Effect of Different Seasons and Explant Types.

Abstract/ Executive Summary:

Considerable effects of different seasons on in vitro cloning of mulberry microshoots were observed. Spring and summer were found to be suitable seasons for maximum rate of shoot proliferation with highest shoot yield. However, maximum shoot length was noticed in the rainy season. Nodal segments were proved to be good explants for micropropagation in mulberry.

Objectives:

To observe whether there is any seasonal influence on in vitro shoot proliferation with an aim to select the best season for explant collection and in Vitro culture on mulberry.

Methodology:

Nodal segments consisting of single node "and shoot apices (ca 1.0-1.5 cm) were coliected from a 10-year old mulberry plant (Morus alba L. cv. MB2) in five different seasons, viz., summer, rainy, autumn, winter and spring. The explants were surface sterilized with O.l% HgCl₂ for 10 mins. followed by rinsing with distilled water 4-5 times and were cultured on MS (Murashige and Skoog 1962) medium supplemented with 2mg/l benzyladenine (BA). The pH of all the medium was adjusted to 5.7 with KOH or HC1, prior to the addition of 0.7% Difco Bacto-agar. Twenty ml of medium was dispensed into each of the culture tubes and autoclaved at 12l°C with 15 Ib pressure per cm² for 15 rniris. All cultures were maintained at a constant temperature of 27±1 °C and 16 hours photpperiod with a light intensity of 2000 lux.

Achievement/Findings:

The greater response of nodal segments over the shoot apices can be explained by the absence of apical dominance and presence of axillary buds at a more advanced stage of development. In apple (Hutchinson 1984) and blackberry (Zimmerman and Broom 1980) nodal segments were also proved to be good explant source of in vitro propagation which has absolutely confirmed the present research findings.
