Estimation of Amino Acids of Sericin Protein in Different Developmental Stages of Multi and Bivoltine Silkworm Races of Bombyx Mori L.

Abstract/ Executive Summary:

The amino acid contents in the middle silkgland of one multivoltine (BSR1-J) and two bivoltine (Dong 34-M and BR-111) silkworm races in different larval developmental stages were studied. Results revealed that amino acid contents of middle silkgland varied among the races and different developmental stages. Highest amount of amino acids was found in bivoltine races than the multivoltine one. Maximum amount of arginine. aspartic acid, glutamic acid, glycine, leucine, lysine, methionine, phenylalanine, proline, serine, threonine and valine were observed in Dong 34-M whereas alanine.cystine, histidine, isoleucine and tyrosine were found in RB-111 on 6th day of 5th instar.

Objectives:

To describe a comparative analysis of expression of sericin in different developmental stages of multivoltine and bivolttine silkworm races.

Methodology:

Disease free larvae of one multivoltine race, BSRI-J and two bivoltine races, RB-111 and Dong 34-M were collected from the Germplasm Bank of Bangladesh Sericulture Research and Training Institute, Rajshahi. Twenty larvae of 3rd and 4th moult and two feeding stages, 3rd day of 4th instar and 3rd and 6th day of 5th instar were taken randomly from each race for the experiment. The middle silkgland was dissected out and washed briefly with ice cold 1.15% KCI and then immediately immersed in 10% trichloroacetic acid for proper fixation of sericin protein and stored at 4°C for 12 hours. The coagulated proteins were collected and washed three times with deionized water. These were further washed with 50% alcohol and absolute alcohol for complete removal of water from the samples, then mixed with ether to remove lipid materials and other particles. Sericin protein (250 mg) was hydrolyzed with 8N HCI in the rate of 1 g of silk protein; 10 ml of 8N HCI in an autoclave at 15 Ibs/in2 for 8 hours. Hydrolyzed samples were then neutralized with Ba (OH)2 and centrifuged at 6000 rpm for 10 minutes. Ammo acid contents were estimated quantitatively following methods of Jayaraman (1985).
