Powdery Mildew Disease of Mulberry Leaf in Bangladesh and Assessment of Changes in Amino Acids and Chlorophyll Contents of Diseased Leaf.

Abstract/ Executive Summary:

Powdery mildew disease incidence of mulberry in different parts of Bangladesh was studied and 56-70% infection was found with 21-41% disease severity. Biochemical studies showed changes in amino acid contents in diseased leaves. Glutamic acid was found to be accumulated in diseased mulberry leaves, where as total chlorophyll, chlorophyll 'a' and chlorophyll 'b' contents were found to be reduced in powdery mildew infected mulberry leaves. Total chlorophyll contents of diseased leaves were also found to be reduced by 50%.

Objectives:

In Bangladesh powdery mildew infection is found to be topped the list of fungal diseases and degraded the normal cocoon production.

Methodology:

To study the incidence of powdery mildew disease, mulberry plots of Bangladesh Sericulture Research and Training Institute, Rajshahi, Ishurdhi in Pabna, Rangpur, Dlnajpur, Konabari in Gazipur, Mainamati in Comilla and Chandraghona in Rangamati district were selected. Disease incidence was studied by counting the healthy and infected leaves and disease severity was recorded according to grading method of Slddaramaiah et al. (1978). To extract the amino acids from healthy and diseased mulberry leaves, leaf extraction in 80% ethyl alcohol was done following the methods of Mahadevan and Sridhar (1982). The extracts were then filtered and the volume of the extract was evaporated to about one fourth over a steam bath and then cooled. The reduced volume was then transferred to a 100 ml volumetric flask and made 100 ml with distilled water- Free amino acids present in the healthy and diseased leaf sample were identified by two dimension paper chromatography. Chlorophyll contents of powdery mildew Infected and healthy mulberry leaves were determined with spectrophotometer according to Mahadevean and Srldhar (1982).

Drastic reduction of chlorophyll a and chlorophyll b was observed in diseased mulberry leaves in the present investigation. Total chlorophyll contents of diseased leaves were also found to be reduced by about 50% as compared to the healthy leaves (Table 3). Neema (1991) found reduction in total chlorophyll as well as chlorophyll a and chlorophyll b content in highly susceptible cultivars of betel vine. Ali et al (1992) reported that total chlorophyll as well as chlorophyll a and chlorophyll b contents per unit leaf area and leaf yield per plant decreased with the increased pathogenesis in case of leaf spot disease in mulberry.
