The experiment was conducted at the Goat and Sheep Farm under Goat and Sheep Production Research Division, Bangladesh Livestock Research Institute (BLRI) from July 2009 to July 2010. The frozen thawed semen was processed in Central Cattle Breeding Station and Dairy Farm (CCBSDF), Savar, Dhaka. The ewes were randomly allocated to breed and condition score with two artificial insemination groups; cervical insemination with fresh semen (single AI) and cervical insemination with frozen-thawed semen (double AI). The estrus of the ewes was observed twice everyday with the aid of a vesectomized rem. Cervical insemination was performed after 12 hours of showing the heat. Insemination with diluted semen was performed using pipette attached with a syringe without needle. For cervical insemination with frozen-thawed semen, double AI at 24 hours and 48 hours were done. A standard, commercially available sheep insemination pipette, designed for use with semen in straws, was used for cervical insemination of ewes with frozen-thawed semen. The sperm concentrations of raw semen were 1.9×109 n 2.20×109 sp/ml and the average volume of semen per ejaculate was 0.85 ml. The conception rate to AI of cervical fresh semen was (52.38%) and double cervical frozen semen was (25.00%). Depending on the present findings, it can be concluded that acceptable conception rate may be obtained with artificial insemination using liquid ram semen.

Artificial insemination leads to improved performance and potential of the nucleus herd and permits coordination of breeding policy on a national basis. Therefore the experiment was conducted to know the conception rate of our native sheep by using fresh liquid semen and frozen thawed semen.

The experiment was conducted at the Goat and Sheep Farm under Goat and Sheep Production Research Division, Bangladesh Livestock Research Institute (BLRI) from July 2009 to July 2010. The frozen thawed semen was processing in Central Cattle Breeding Station and Dairy Farm (CCBSDF), Savar, Dhaka. The ewes were randomly allocated by breed and condition score with two artificial insemination groups; (1) Cervical insemination with fresh semen (single AI); (2) Cervical insemination with frozen-thawed semen (double AI). Semen was collected from the same three rams of Noakhali, Tangail, and Noagaon region at BLRI sheep farm. For the frozen semen, the semen was collected, evaluated, processed and frozen in 0.25 ml straws according to their standard protocols. The ram was allowed to mount a ewe and the penis was manually diverted into the artificial vagina by grasping the sheath without touching the penis. When ejaculation has occurred, the artificial vagina was removed and immediately after collection semen was kept in warm water at about 41? until further processing. Sperm concentration was measured by haemocytometer method (Herman and Madden, 1963)directly.The pH of semen was measured directly with the aid of nitragene paper while the individual motility live, dead sperm and normal, abnormal sperm (fig. 1 and 2) were observed under microscope. One step tris based, egg yolk and glycerin based extender used for semen processing. Citric acid, de fructose and antibiotics also used for extender (each 100 ml diluter contained 20 ml egg-yolk, 80 ml 2.94% sodium-citrate buffer, penicillin @ 1000 IU/ml, streptomycin @ 1000 mg/ml, 6 mg/ml sulphanilamide powder). Essentially, only ejaculates with >80% progressive motility and 80-100×106 motile sperm/ml were used and were diluted to achieve around 300 million sperm/breeding unit. In addition, after freezing and preserve in liquid nitrogen for 48 hours and than thawing the straw and evaluate the sperm motility by trinocular phase contract microscope with monitor and found 45-50% forward movement which were accepted for use. The straws from each ejaculate from the individual rams were pooled prior to use. The estrus of the ewes was observed twice everyday with the aid of a vesectomized ram and inseminated after 12 hours from the onset of heat. The ewes that were in heat kept in pen separately. Cervical insemination was performed after 12 hours of showing the heat. Insemination with diluted semen was performed using pipette attached with a syringe without needle. Before the semen was expelled out of the insemination pipette, the speculum was removed to avoid pneumovagina and subsequent discharge of semen. For cervical insemination of frozen-thawed semen double AI at 24 hours and 48 hours were done. A standard, commercially available sheep insemination pipette, designed for use with semen in straws, was used for cervical insemination of ewes with frozen-thawed semen. The straws were thawed immediately prior to use by placing them in a water bath at 37? for around 10 sec. They were then dried, one end was cut off and the straw loaded into the AI pipette. The same three rams were also used to supply fresh semen for ewes in group (i). Ejaculates (3 per ram) were collected, checked for progressive motility, pooled and diluted with Egg-yolk-citrate to achieve a final concentration of 300×106 motile sperm/0.25ml. A conventional Perspex insemination pipette, coupled to a syringe with rubber tubing was used to facilitate deposition of 0.25 diluted semen into the posterior or of the cervix. Assumption conception rate was calculated on the basis of the ewes not coming into heat again within forty days. Data were treated and statistically analyzed by using Statistical Package for Social Science (SPSS, version 11.5). Mean comparisons were made using Duncan multiple range test (DMRT).

Depending on the present findings, it can be concluded that acceptable conception rate may be obtained with artificial insemination using liquid ram semen. However, further study should be required on intrauterine insemination with frozen-thawed semen for acceptable conception rate.
