Detection of Antimicrobial Drug and Feed Additives Residue in Food from Poultry Origin (meat and Eggs).

Abstract/ Executive Summary:

Chicken (50 birds / division) and eggs (30 egg / division) were collected from different markets of Dhaka, Rajshahi, Sylhet, Barisal and Chittagong City Corporation. Breast muscles, liver & Kidney and eggs of chickens were taken to test the residue level. Both the tissue and egg samples were tested with commercial gentamicin ELISA kits. Out of these 41% tissue samples and 47% egg samples were found to contain gentamicin residue above maximum residue limit (MRL). In this study, highest mean of gentamicin residue was found in egg yolk and in case of tissue samples mean residue in kidney was higher than other organs followed by liver and breast muscles. All these results showed that high and violative levels for gentamicin residues.

Objectives:

a. To detect antimicrobial drug residue in food of poultry origin like, meat and egg. b. To identify a locally suited, economic but sensitive detection method.

Methodology:

Collection and transportation of samples: Chicken and eggs were collected from different markets of Dhaka, Rajshahi, Sylhet, Barisal and Chittagong City Corporation. Breast and thigh muscles, liver & Kidney of broiler chickens were taken as tissue samples. Collected tissue samples were transferred to the laboratory taking necessary safety measures and maintaining cold chain properly. Eggs and tissue samples were stored - 4? and at -20? respectively until of analysis. 2. Sample preparation: (a) Tissue samples: Samples were processed in the laboratory according to the instructions of the ELISA kit. Approximately 50 gm of each tissue sample was homogenized separately with tissue homogenizer. 1.0±0.05 g homogenate was taken into a 50 ml polystyrene centrifuge tube and 5 ml 3% trichloroacetic acid solution was added. Then the mixture shacked for 1 min with shaker to mix the solution completely and was centrifuged for 5 min @ 2000g. Transferred 200 ml of the supernatant liquid to a 2 ml polystyrene centrifuge tube and then added 200 ml 1X sample extraction buffer and pH adjusted 7.4 with 1M Na0H. 50 ml of the prepared solution was used for assay. (b) Egg samples: Homogenized egg (both egg white and yolk) and took 0.5 g in a 15 ml polystyrene centrifuge tube. 1X sample extraction buffer was added with this and mixed well by vortexing. The sample was centrifuge for 5 min @ 2000 g. Transferred 1 ml of the supernatant and 50 ml of the prepared solution was used for assay. 3. Procedures for ELISA test: All reagents were taken out at room temperature (20-25?) for more than 30 min, 50 µl standard sample solution or prepared samples were added the to corresponding wells. 100µ antibody # 1 was added in each well. Then mixed gently by shaking the plate manually and incubated for 30 min at 25? with a cover. After 30 min the cover was removed and poured the liquid out of the wells and rinsed the microwells with 250 µl of 1X diluted wash solution for 4 times. Residual water was absorbed with absorbent paper. The 1X antibody # 2 solutions was added @ 150 µl to each well and mixed gently by shaking the plate manually and incubated for 30 min at 25? with a cover. Wash step was repeated again. 150 µl TMB substrate added to each well and incubates for 15 min at 25? with cover. Finally 100 µl stop solutios was added to each well and mixed gently by shaking the plate manually. The absorbance was measured at 450 nm against an air blank within 5 min after addition of stop solution.

Achievement/Findings:

In this study, highest mean of gentamicin residue was found in egg yolk and in case of tissue samples mean residue in kidney was higher than other organs followed by liver and breast muscles. Through these levels gentamicin residues are high and violative levels for human health but this was not unusual since the liver and the kidney are the major storage and excretory organs for gentamicin and are parenchymatous in nature.
