Surveillance of Zoonotic Influenza Viruses Hosted by the Swine Population of Bangladesh.

Abstract/ Executive Summary:

A total of 240 pig sera samples were collected from Dinajpur district and slaughter houses of Gazipur district. Out of these, 184 sera samples were subjected to ELISA test against HINI and H3N2 antibodies. All these samples were also subjected to HI test for H5 and H9. Seroprevalance of swine influenza HINI and H3N2 in pigs were 79% and 53% respectively. Combination of both subtypes was deteted in 44% pigs. Whereas in HI test, 3% samples were sero-positive for H9 and combination of HIN, H3N2 & H9 was also found in 3% pigs. It’s assumed that frequently consecutive or mixed infection of influenza viruses in of Bangladesh may provide an opportunity for genetic reassortment of these viruses.

Objectives:

In Bangladesh yet no surveillance is done on circulating influenza viruses in the swine population. The health and socio-economic condition of swine rearer is not good and are not aware about personal as well as swine health. So there is a chance of transmission of zoonotic influenza viruses from human to swine and swine to human as well as re-assortment. In order to assess implication for human health, it is important to know whether the reported infections in pig are rare events in the swine population of Bangladesh. Considering the above facts the present study was designed for sero-surveillance of currently prevail zoonotic influenza viruses in swine population Bangladesh.

Methodology:

Collection and transportation of samples: Blood samples were collected from jugular furrow of live pigs. In the slaughter house, blood samples were collected with onetime used plastic glass from the puncher area (heart) from where externally bleeding during slaughtering of pigs. 2. Processing and transportation of samples: Collected blood samples were centrifuge for 5 min @ 1500 g and serum were collected in 1.5 ml eppendorf tube. Sera samples were transferred to the laboratory taking necessary safety measures and maintaining cold chain properly and stored at -20? until of analysis. 3. Procedures for ELISA test: (a) Wash solution preparation: The wash concentrate must be allowed to reach room temperature (20-25?) and mixed the concentrate in the bottle by inversion to dissolve any precipitated salts and to ensure a homogenous solution. The wash concentrate diluted 10-fold (1/10) with distilled water. (b) Preparation of serum: Sera were diluted 40-fold (1/40) in sample diluents. The position of each diluted specimen in the dilution rack and the corresponding wells of the antigen-coated plate recorded properly. (3) Assay Protocol: All reagents must be allowed to reach room temperature (20-25?) before use. Reagents should be mixed by gentle swirling or vortexing. Undiluted negative control and undiluted positive control were dispensed (100 µl) into wells A1, B1 and C1 & D1 respectively. 100 µl of each diluted serum specimen dispensed into the applicable well. The plate was incubated at room temperature for 30 min. The liquid contents of all wells were aspirated and washed all wells with approximately 300 µl each of IX wash solution. Conjugate was dispensed @ 100µl each well and incubated at room temperature for 30 min. Again all wells washed with approximately 300 µl each of IX wash solution and dispensed 100µl substrate solution into each well. The plate was incubated at room temperature for 15 min and finally 100 µl stop solution was added into each well. The OD value was measured and recorded at wavelength of 650 nm using spectrophotometer.
