Assessment of Leaf Spot Disease of Mulberry (morus Sp.)leaf in Bangladesh and Change in Amino Acids Content of Diseased Leaf.

Abstract/ Executive Summary:

Leaf spot disease incidence of mulberry in different parts of Bangladesh was studied and 55.43 to 69.00% infection was found with 20.17 to 40.26% disease severity. Biochemical studies showed changes in amino acids content in the diseased leaves and eight amino acids were found in the healthy leaves and five amino acids were found in the infected leaves. Amino acids were always found to be higher in the leaves than the infected ones with total absence of Lysine, Aspartic acid and Alanine. Leaf spot infection reduced Serine, Iso-Leucine, Leucine and Glutamic acid by 50%, 33.33%, 50% and 33.33% respectively.

Objectives:

To study leaf spot disease of mulberry leaf and change of amino acid content of diseased leaf.

Methodology:

To study the incidence of leaf spot disease eight nurseries under Bangladesh Sericulture Board were selected in different districts of Bangladesh. These were Rajshahi, Pabna, Bogra, Rangpur, Dinajpur, Gazipur, Comilla and Rangamati districts. Disease incidence was records during the months of July to September in 2000, 2001, and 2002.Disease incidence was studied by counting the healthy ad infected leaves in three vigorous branches of eafh plant and diseases severity was recorded according to grading metod of Siddaramiah er al. (1978) as follows: 0= No infection (healthy), I= 1-5%, II= 6-25%, III= 26-50% IV=51-75%, V= 76-100%. Disease severity in terms of disease index (DI) was calculated by the formula: Sum of numerical value x 100 DI. Total no. of leaves graded x maximum grading in 5 Where numerical values were obtained by multiplying the number of leaves with their respective grading. All the studies were made in there replications and average results were calculated. The DI values were transferred into angular values and statistical analysis was completed. For biochemical study amino acids were extracted from healthy and diseased mulberry leaves. Leaf extraction in 80% ethyl alcohol was done following the methods of Mahadevan and Sridhar (1982). The extracts were then filtered and the volume of the extract was evaporated to about the volume of the extract was evaporated to about one fourth over a steam bath and then cooled. The reduced volume was them transferred to a 100 ml volumetric flask and made 100 ml with distilled water. Free amino acid present in the healthy and diseases leaf sample were identified by two dimension paper chromatography.

Present study indicates that due to infection and change in biochemical contents the metabolic process ofthe infected plant is altered as compared to the healthy ones which leads to cause yield and quality reduction of mulberry leaves.
