The study was conducted at Molecular Breeding Laboratory, BARI, Gazipur with two onion varieties named BARI Onion-1 and BARI Onion-5 during Rabi season of 2012 to observe the changes of the levels of Glutathione-S-transferase (GST) and Glyoxalase-1 (Gly-1) and to observe the physiological basis of changes during storage of onion. Data were recorded on the specific activity of GSTs and Gly-1 on seven different date of preservation viz. 21 October, 06 November, 21 November, 06 December, 21 December, 06 January and 21 January. The results showed that the highest (548.2772 nmol/min/mg/protein) GSTs were recorded in case of BARI Onion-1 on 21 November and the lowest (404.5986 nmol/min/mg/protein) on 06 November. In case of BARI Onion-5, it was recorded the highest (3769.3630 nmol/min/mg/protein) on 06 January and the lowest (3262.3600 nmol/min/mg/protein) on 21 January. On the other hand, the highest (7.3939 nmol/min/mg/protein) Gly-1 was recorded on 21 January in case of BARI Onion-5 and the lowest (3.0850 nmol/min/mg/protein) on 06 November. Certain changes indicate that there might have some incident of accumulation of toxin. Some fungal infection was found. There might also be accumulated methyl oxal, which might change the activity of GSTs and Gly-1.

to check the variation of enzyme activity level over time of storage in BARI Onion-1 and BARI Onion-5
The study has been conducted at Molecular Breeding Laboratory, BARI, Gazipur. Two varieties of onion viz., BARI Onion-1 and BARI Onion-5 were used as planting materials which was collected from Regional Spices Research Centre, Gazipur. Data were recorded on assay activity of enzyme, measurement of GST: (Glutathione-S-transferase), Gly-1 (Glyoxalase-1) and protein concentration on seven different date of preservation viz. 21 October, 06 November, 21 November, 06 December, 21 December, 06 January and 21 January.

The drastic change of some enzyme i.e., the increase in activity of GSTs in BARI Onion-5, 3769.3630nmol/min/mg/prtoein on 06 January and 3262.3600nmol/min/mg/prtoein on 21 January and Gly-1, 7.393922nmol/min/mg/prtoein on 21 January was found which indicate that there might have some incident of accumulation of toxin. Among them, pathogen inoculation might be one of the causes, accordingly some fungal infection was found. There might also be accumulated methyl oxal, which might change the activity of GSTs and Gly-1.
