Establishment of in Vitro Embryo Production System with Slaughterhouse Ovaries

Abstract/ Executive Summary:

This study was conducted to provide basic biometrical information on different organs of normal female genital tract and follicular dynamics of indigenous cattle. About 80 genital tracts of adult non-gravid female cattle were collected from local abattoir in physiological saline at 25-30⁰C and transported to the laboratory within 4 to 5 hr of slaughter. Dimensions of oviduct, uterine horn, uterine body, cervix and ovary were measured using standard protocol. Moreover, the follicles numbers on the ovarian surfaces, oocyte aspiration rates and morphological quality of aspirated oocytes were evaluated in this study. The genital tracts were classified in to two groups (cows: delivered minimum one calf vs. heifer:≥30 months age) for measurement of dimensions. The ovaries were picked up from the genital tracts and grouped on the basis of corpus luteum (CL) ovary with rudimentary CL; ovary with active CL and ovary without CL and ovarian position in the tract (right and left ovaries). Each ovary was weighed placing on an electric balance and dimensions were measured with vernier calipers. All visible follicles were counted and graded as small (≤3.00mm), medium (3.0 to 8.0 mm) and large (≥8.0mm). The oocytes from each ovary were aspirated using 10 mL syringe attached with a 21G needle. The aspirated material was poured onto a 35 mm petridish and the number of oocytes per ovary was recorded under a microscope at low magnification (4x). The oocytes were graded into four groups considering their cumulus cell attachment and cytoplamic appearance. Data were analyzed using ANOVA following principles of completely randomized design experiment. DUNCUN multiple range test was used to compare mean differences in appropriate cases. Results showed that the oviduct, horn and body of the uterus and cervix of cows genital tract were longer and wider than heifers. Moreover, the length of ovary varied with ovarian position and presence or absence of corpus luteum. However, ovarian position or presence/absence of CL did not determine the width of ovary. The weight of ovary did not differ between left and right. However, in presence of CL, the ovarian weight was increased. Each ovary possessed 9.65±5.91 visible follicles belonging to small, medium and large groups, respectively. Neither ovarian position position nor presence of CL has effect on follicle numbers in the ovary. Number of oocytes aspirated from an ovary and oocyte recovery rates were independent of ovarian position and presence of CL. About 52% of the aspirated oocytes were morphologically cultivable. The present study provides baseline information on biometry of different organs of female genital tract and follicular dynamics of indigenous cattle.

Objectives:

Methodology:

Laboratory preparation: Before collection of female genital tracts all logistics including a compound microscope, a vernier scale, a metric ruler, an electronic balance, a large dissection tray, culture dishes, Pasteur pipette, physiological saline, DPBS, hand gloves and other necessary materials were collected and placed in to the working laboratory. Collection of cow genital tracts: About hundred non-gravid genital tracts were collected from slaughtered cows and heifer from abattoir located at Mohammadpur, Dhanmondi, Dhaka. Biometrical measurement: Examination of the specimens was carried out under bright light. All measurements were recorded in centimeters/millimeters and all weights in grams. All measurement were done according to Gupta et al (2011). Statistical analysis: All measurements were recorded in a tabular form. The tabulated data were entered into computer using Microsoft excel worksheet and the data were transferred into SPSS data file for analysis of descriptive statistics.

Achievement/Findings:

In summary, it was observed that the length and width of oviduct, uterus and cervix of cows are greater compared to non-pregnant heifers. Ovarian length is varied with ovarian position and corpus luteum. The weight of ovary was also influenced by corpus luteum. Number of follicles in an ovary, their recovery rate and quality of recovered oocytes may be cultured for in vitro embryo production.
