Factors Causing Emerging Shrimp Diseases and Development of Their Health Management Strategies

Abstract/ Executive Summary:

A total number of 79 samples of Tiger shrimp brood were tested in PCR laboratory of Marine Fisheries and Technology Station, Cox’s Bazar. It was found that in case of tiger shrimp brood highest WSSV prevalence was found 80% in the month of April and lowest WSSV prevalence was found 60% in the month of January. Average WSSV infection in brood tiger shrimp was found 73.4%. From the data of nauplii, it was found that highest WSSV prevalence was 55.3% in the months of April and lowest was 36% in the month of January. Average WSSV infection in tiger shrimp nauplii was found 47.3%. From the PCR tested data of PL, it was found that highest WSSV prevalence was 22.2% in the month of April and lowest was 0% in the month of January, February and March. Average WSSV infection in tiger shrimp post larvae was found 8.9%. Samples of tiger shrimp post larvae and waters were collected from two hatcheries and plated onto agar plates containing thiosulphate citrate bile salt (TCBS) agar medium. The study revealed that the Vibrios densities was below lethal level as mentioned Vibrios lethal dose for Penaeus monodon post larvae was 2.6 x 103 cfu/ml.

Objectives:

Assessment of the present status and seasonal variation of WSSV infection in shrimp using PCR technique and Diagnosis of bacterial diseases of shrimps in hatchery

Methodology:

Shrimp (P. monodon) mother broodstock were collected from fishing trawler (sea) and from different hatcheries. A portion of pleopod was collected from broodstock without sacrificing the animals and will be taken into Eppendorf tube containing 70% alcohol. The shrimp nauplii, post larvae (50-60 nos) was collected from hatcheries and was put into zipper poly bag or into Eppendorf tube. Immediately after collection, the samples was transported to the laboratory for analysis. Cultured shrimp was collected from shrimp ponds/ghers. Standard bacteriological culture technique (sterilization, isolation, incubation etc.) was followed to culture and identify bacterial diseases. Tiger shrimp brood (a tip of pleopod, after spawning), nauplii, shrimp larvae (Early PL, Mid PL and PL-15) were collected from three hatcheries. Tiger shrimp DNA was extracted according to the protocol of IQ 2000 WSSV detection and prevention system. For WSSV detection nested PCR method was used following the PCR protocol of IQ 2000 WSSV detection and prevention system at the laboratory of Marine Fisheries and Technology Station, Cox’s Bazar.

Achievement/Findings:

Healthy and virus free broodstock is the key to get good quality post larvae and virus free post larvae are the key to fetch successful shrimp production. Before stocking to the culture ponds/ghers it is important to test shrimp PL by PCR and only WSSV negative PL should be stocked to avoid risks. PCR test can reduce the catastrophic losses from WSSV.
