Screening of Rice Genotypes at High Salinity Stress of the Seedling Stage of Rice in Boro Season

Abstract/ Executive Summary:

An experiment was conducted in the Plant Physiology Division, Bangladesh Rice Research Institute, Gazipur during Boro season, 2011 to identify advance breeding lines of rice genotypes having salt tolerance at seedling stage. Forty three advance breeding lines were tested at 12ds/m. Eight advance breeding lines showed visual score 3 to 5 with more than 70% survivability. Based on the visual score and survivability, 8 genotypes out of 43 were found tolerant in comparison with checks.

Objectives:

To identify salt tolerant advance breeding materials at seedling stage of rice.

Methodology:

Sprouted seeds were sown on nylon net fitted with styro-foam floating on Yoshida,s full strength culture solution. Salinity stress @ 12 dS/m was applied 6 days after sowing. The culture solution was changed once a week throughout the experimental period. The pH and EC of the culture solution was checked daily to maintain at 5.0 and 12 dS/m respectively. In each tray FL478 and IR29 were used as a tolerant and susceptible check, respectively. Forty three genotypes were considered for the study. When susceptible check IR29 died, visual score was made according to SES system and living plants were recorded. The scoring system for screening at seedling stage as follows: Score Observation Remarks 1 Normal growth, no leaf symptoms Highly tolerant 3 Nearly normal growth, but leaf tips or few leaves whitish and rolled Tolerant 5 Growth severely retarded; most leaves rolled; only a few are elongating Moderately tolerant 7 Complete cessation of growth; most leaves dry; some plants drying Moderately susceptible 9 Almost all plants dead or drying susceptible Based on above scoring system the breeding lines having scored 1 to 5 were treated as tolerant and scored 7 to 9 were treated as a susceptible.

Achievement/Findings:

Among 43 genotypes, only 8 (Eight) genotype shows visual score 3 to 5 that was tolerant to moderately tolerant which were IR45427-2B-2-2B-1-1, IR76393-2B-7-1-1-3-1, BR7105-4R-2, BRRI dhan47, IR83416-7-B-12-3-1-3-AJYI-B, IR87888-3-AJYI-B, IR09T484, IR84089-7-3-AJY1-B. The survivability of these genotypes varies from 70% to 90%. However, other genotypes shows visual score 7 to 9 with 0 to 70% survivability that was susceptible to highly susceptible. The tolerant check FL478 showed the tolerance score 3 with 100% survivability. On the other hand the susceptible check IR29 showed the tolerance scores 9. None of the plants of IR29 could survive.
