Effect of Different Carbon Sources on Mycelial Growth and Sclerotia Formation of Rhizoctonia Oryzae Sativae.

Abstract/ Executive Summary:

Among six different Carbon sources used in this experiment, the best mycelial growth found in Dextrose (0.43mm/ hr) followed by Sucrose (0.42mm/hr). The most unsuitable source is Fructose (0.26mm/ hr) followed by Xylose (0.35mm/hr). In case of duration of complete mycelium running, the minimum time was recorded in Dextrose (105 hrs.) Maximum time was found in fructose (172 hrs.) followed by Xylose (130 hrs.). The highest sclerotial formation was found in Dextrose

Objectives:

To know the effect of Carbon Sources on mycelial growth and sclerotia formation.

Methodology:

Different carbon source such as Dextrose, Sucrose, Galactose, Xylose, Fructose and Lactose mixed separately with 2% agar and 100 ml potato extract for medium preparation. pH ranges were taken. Mycelial disks of 6mm diameter, were transferred from the margin of 4-day-old colonies at the centre of PDA plates and incubated at 27±2°C. Four replicate plates were used for each treatment. Redial mycelial growth and sclerotial data were taken. Carbon source, Dextrose (pH 6.7), Sucrose (6.8), Galactose (6.8), Xylose (6.6), Fructose (6.7) and Lactose (6.8) were used in this experiment.
