In Vitro Regeneration of Six Jute Genotype in Bangladesh

Abstract/ Executive Summary:

Six variety of jute (Corchorus capsularis L.) were raised in this experiment to observe their regeneration potentiality and to establish a suitable in vitro plantlet regeneration protocol of jute. Cotyledons were used as explants. MS medium supplemented with different phytohormone concentrations and combinations were used to observe the callus induction ability of the explants. The highest callus induction (83.89%) was observed in CVE-3 on MS medium supplemented with 2 mg/L BAP + 0.5 mg/L IAA. Calli ere maintained in the same medium for plant reneration. The highest percentage of regenerants were obtained from CVE-3 (86.87%) followed by BJC-7370 (74.64%). Among the phytohormone combinations, MS medium supplemented with 2 mg/L BAP + 0.5 mg/L IAA showed the highest shoot regeneration (84.13%). Root formation from the regenerants was best on half strength MS medium supplemented with 0.6 mg/L IBA. In vitro regenerated plantlets were successfully established in the field.

Objectives:

The objectives of this experiment was

1. Optimization of in vitro regeneration performance of . varieties.

2. Standardization of a protocol for callus induction and shoot regeneration with different hormonal treatments. and

3. Screening of some genotype of Corchorus capsularis L. for their tissue culture potentiality.

Methodology:

Experimental materials : Healthy seeds of six varieties of Corchorus capsularis L. (CVL-1, CVE-3, BJC-7370, C-83, CC-45 and D-154) were used as experimental materials were sterilized before use. Media: The hypocotys and cotyledonary nodal segments (cotyledons) from seedlings raised in axenic culture were used as the source of explant. the explants were first cultultured on agar solidified MS medium supplemented with 1.0, 2.0, 3.0, 4.0 and 5.0 mg/L BAP in combination with of 0.5 mg/L IAA for callus induction and shoot regeneration. About 2-3 cm in long shoots were sub cultured in vials with freshy prepared medium containing half strength MS medium supplemented with 0.2, 0.4 and 0.6 mg/L IBA for root formation. Culture techniques: Sterilized seeds were placed into sterilized seed germination medium in culture vessels. Twenty five seeds were then placed in each vials. The culture was then incubated in dark till the germination of seeds. These were transferred to 16 hours light for normal seedlings growth. Seven days old seedlings were used as sourch of contamination-free explants. The seedlings raised in axenic culture were used as source of different kinds of explants. The hypocotyls and cotyledonary nodal segments (cotyledons) were used as explants. Attempts have been taken for the induction of embryogenesis and organogenesis using different explants in MS medium supplemented with different hormones. After the calli attained a size of about 20-25 mm in diameter, they removed aseptically and sub-cultured to other vials, the calli were again cultured on freshy prepared medium containing the same hormonal supplements, for shoot induction from the cells. When shoots were about 2-3 cm in length, they were rescued from the vials aseptically inside the laminar air flow cabinet, and were again cultured in vials with freshy prepared root induction medium to form root.

Among the six varieties of jute (Corchorus capsularis L.) CVE-3 performed best and this finding will be helpful in different biotechnological approaches.
