An experiment on Potato virus Y (PVY) isolation and multiplication was conducted at TCRC Insect proof net-house during 2010-11 cropping season. PVY symptomatic potato samples were collected from field and inoculated mechanically into the indicator hosts. PVY produced local lesion into the indicator host and was multiplied into the propagation hosts like, Nicotiana tabacum var. White Burley and Samsun and in Nicotiana glutinosa for purification. Leaf samples were preserved in the refrigerator and will be used for purification through differential centrifugation.

To purify potato viruses in order to produce antisera for indexing diseases.
A set of indicator plant such as Nocotiana glutinosa L., Nicotiana tabacum L. var. White Burley and Samsun, N. benthamiana L., Physalis floridana L., Chenopodium amaranticolor L., C. quinoa L., Datura metel L., D. Stramonium L., Gomphrena globosa L., Lycopersicon esculentum (Rotzer Tomato) were planted at insect free net house of TCRC, Joydebpur, Gazipur for isolation of major potato viruses through mechanical inoculation. PVY strains were identified through host range studies. Mixed infected plants with PVY and PVX were separated by inoculating into Datura stramonium which is immune to PVY and susceptible to PVX. PVY isolate was maintained in their local lesion host C. amaranticolor and was multiplied into the propagation hosts like, N. tabacum var. White Burley and Samsun and N. glutinosa. The multiplied virus samples were kept in the refrigerator and will be used for purification through differential centrifugation in due course.
