Cultures of Anthers of Cultivated Varieties of Kenaf, Mesta and Jute

Abstract/ Executive Summary:

Anthers of four species C. Capsularis (Var. CVE-3), C . olitorius (Var. O-4), H. cannabinus (Var. HC-2) . H. Sabdariffa (Var. HS-24) were used for anther culture with the aim to be used for haploid production. Anthers were collected from the matured plants of all varieties and were surface sterilized with 0.1% HgCl₂.The anthers with appropriate size (Uninucleate stage) wre collected and cultured in 100 ml flasks on agar solidified MS medium supplemented by IAA and BAP. Calliwre produced from the anthers of all species three to four weeks after culture. Calli of C. capsularis became green compact and light yellow and compact calli were observed from the anthers of C. olitorius. However, plant regenerationwas not obtained from any of the anther-derived calli.

Objectives:

1.Production of genetic variants of C. capsularis and C. olitorius, kenaf and mesta through in vitro culture of explants and their utilization in breeding programme for varietal improvement.

2. Production of haploid plants through anther culture.

Methodology:

1. The flower buds (when the pollen is at uninucleate stage) would be chosen for anther culture 2. Flower buds would be sterilized by using ethanol (2 minutes) and then by HgCl₂ and then by rinsing in sterile water 3-4 times 3. Removal of anthers from the flower buds in sterilized condition and culture on MS agar solidified medium containing auxins and cytokinins in various combinations 4. Initially the cultures would be kept both in light or in dark at 28° c . After callus induction the cultures would be subcultured on the same medium as well as with other combinations of hormone until plant regeneration is obtained. The cultures should be kept under 12h photoperiod 5. Each experiment should be repeated 3 times using minimum of 3 replicates.
