Optimization of Culture Conditions for Plant Regeneration from Explants of Kenaf.

Abstract/ Executive Summary:

Seeds of kenaf(Var. HC-2) and one variety were surface sterilized by 0.1% HgCl2 followed by several washing with sterilized tap water. Seeds were germinated on the surface of 80 ml of agar solidified MS medium in 500 ml flasks. Fifteen day old seedlings were used for in vitro culture. Cotyledons were excised from the in vitro grown seedlings and were cultured in 250 ml conical flasks on the surface of 70 ml MS agar solidified medium with various concentrations of IAA or NAA and BAP. The differences within treatments were found highly significant (p<0.001). Hormone combination IAA(0.5 mg/L) + BAP (5 mg/L) produced the highest percentage of shoots (59.5%). This was found significantly higher than other combinations. However in case of NAA (0.5 mg/L) + BAP (5.0 mg/L) the differences was not significant. IAA 0.5 + BAP 3.0 mg/L produced the lowest percentage of shoot (25.00%). The overall differences among the treatments were found significant (p<0.050. The highest number of shoot was produced with the addition of IAA 0.5 and BAP 5.0 mg/L and the lowest number of shoot was produced with IAA 0.5 and +BAP 3.0 mg/L

Objectives:

The aim of this experiment was to establish a plant regeneration protocol from the explants of kenaf to be used for the production of somaclonal variants and to be used for gene insertion in future.

Methodology:

a) Cotyledon segments, hypocotyle segments, root segments, nodal segments, shoot apices and cotyledons (with attached petioles) of mesta would be cultured in flasks/jars, on the surface of 50 ml of agar solidified MS medium supplymented by various combinations of auxins and cytokynins b) Optimization of culture conditions eg. temperature and intensity of light and photoperiod. c) Regular subculture of the regenerated materials and development of root induction from the cultured shoots. d) Transfer of the regenerated plant materials into pots containing soil. e) Record of morphological changes in the regenerated(potted and in vitro) plants.
