Mycorrhizal association is the most essential component of soil environment for making soil a living entity where soil, fungal structure and plant root collectively form a close beneficial relationship. Comparative survey of status of AM fungus in different AEZ soil with colonization in different legumes and non-legume crops and presence of AM spores were done earlier at Microbiology Laboratory during the year of 1998 to 2001 under ARM Project and being continued. Plant and soil samples were collected from four sampling sites such as BARI central farm, Gazipur, RARS Jamalpur, RARS Ishurdi and RARS Hathazari, Chittagong. Out of 325 soil root mixed sample, a total of 48 mixed dense source of AMF found effective in trapped culture and are being continuously multiplied. Another collection of five mono culture of mycorrhiza were collected from ICRISAT, India. These inoculum sources are being maintained round the year and being used as the inoculum source for all mycorrhizal experiments of the soil science division of BARI.

01 Identification, classification and characterization of indigenous genera of mycorrhiza from different AEZ

2. To carry out further study using the materials received from the existing ARMP Mycorrhiza project.

3. To carry out greenhouse study with the existing trapped mixed source of mycorrhiza to continue their generation.

4.To use the mixed mycorrhiza as a source of pure mono culture production.

5. Ultimate goal is to get suitable inoculum for legume and other crops.

Potential inoculum production of mycorrhiza needs an interrelated chain of study. The whole process has been stepped down for practical use as follows : 1. Soil-root samples of different legume and non legume crops from different Agro Ecological Zones were collected. Roots and spore were separated and studied for mycorrhizal colony, spore separation and identification. 2. Separate pot culture study was conducted from roots and spores. Four methods of pot culture were carried out such as culture from intact soil core, culture from separated spores, culture from root segments and sub pot culture from existing pot culture. 3. Routine sub-sampling was done from pot culture and initiation of new pot culture with small cores to get a purified monoculture source on mycorrhizal inoculum. 4. Round the year maintenance, multiplication and preservation of mixed Mycorrhizal source with suitable living host trap crop (maize, sorghum, cowpea and soybean) for target inoculum production. Sample collection: The above ground portion of the crop was removed and nearly 15-20 cm deep X 12 cm diameter soil with roots of the target plant was taken carefully. Preparation of pot culture media: Pot cultures were conducted using sand: soil (2:1) mixture with prior sterilization. Management of the pot culture: Initially the pot culture was watered with standard nutrient solution having P level adjusted to approximately 60 % of standard requirement. Moisture condition of the pot culture was maintained at their field capacity. Plants were harvested at 50-60 days after planting. Half of the roots were separated for microscopic study and remaining were chopped and mixed with the soil to grow plant for the next generation.

A total of 53 isolated mixed local sources and 5 monosporic culture of AM inoculum obtained from ICRISAT, India are being maintained and multiplied continuously. All the sources of inoculum are maintained properly. Mixed mycorrhizal inocula JL2000, JD2000, ISD2000, HT2000 and ICR2000.coleced from Jamalpur, Joydebpur, Ishurdi, Hathazari and ICRISAT, India respectively were used as inoculum source in the experiments. There are three cycles of trap crop for the entire season of 2003-2004. Sorghum, onion and maize were used as tarp crop. The range for percent root colonization is 62% to 100% for all trap crops. Soils sieved were examined for spore number and spore number / 100 g are maintained at a range from 136-498 no. / 100 g of soil.
