Isolation and Characterization of Newer Lignocellulolytic Microorganisms and Assessment of Their Enzyme Activity.

Abstract/ Executive Summary:

Considering the enzyme activity,both xylanase and CM Case, none of the four isolates could be claimed as highly potential ones.However, these are simply important because they are able to grow at 55 0 C both in liquid as well as in solid agar medium and can be named as thermophilic strains. The morphological , physiological and biochemical characteristics of these 4 strains will be studied and their identification will be tried for their proper use.

Objectives:

To find out potential strains of microorganisms capable of producing highly active lignocellulolytic enzymes that can be used for the quality improvement of jute fibre, fabrics and other products.

Methodology:

Lignocellulosic decomposed samples were collected from various places. As usual dilution plat culture technique was followed for primary isolation of the selective fungal colonies. Before going to make dilution of the samples, they were cut into small pieces, put into sterile water in conical flasks, stirred to mix well and were kept at 80 0 C for several hours. This was done to reduce the microbial population growing at lower temperature ( 40 0 C ). The basal medium for plate culture contained yeast extract 0.5%,KH₂PO₄ 0.05 % , NH₄)2SO₄ 0.5% CaCl₂ 0.005% ,Carbon source ( MCC / Xylan ) 2.0% and pH 6.0. The culture plates were incubated at 40 0 C and 55 0 C simultaneously for 3 to 7 days. Moderate to vigorously colonies growing on the medium were isolated and purified through successive transfer and subcultures.

Achievement/Findings:

Initially 96 fungal colonies were isolated and purified. Secondary selection from among these 96 colonies was done by growing them at 55 0 C on agar medium containing exclusively microcrystalline cellulose as sole source of carbon along with other ingredients. Out of these 96 isolates only nine grew profusely on agar medium showing distinct hydrolyzing zone. From these nine isolates, 4 were finally selected for carrying out shake culture fermentation to ensure production of extracellular cellulase and xylanase by them in liquid medium.
