Treatment of the Crude Pulps by Xylanase / Ligninase to Supplement Chemical Bleaching and Making of Hand Sheet Papers

Abstract/ Executive Summary:

Enhanced brightness of Soda-Aq. pulp and Kraft pulp was obtained, but not up to the expected level .The effectiveness of xylanase pretreatment as vthe release of chromatophore materials, carbohydrates and reducing sugars from pulps as well as the modification of pulp properties such as kappa number, brightness and viscosity.

Objectives:

Selection , production and application of microbial enzymes for bleaching of jute / kenaf pulps through biopulping process. Reduce the production cost of pulp and paper. Cooking chemical used as enzyme for bleaching and improving pulp properties.

Methodology:

Production of xylanase : Xylanase was produced by using a thermophilic fungus, Thermomyces lanuginosus ( RT -9 ) in solid state fermentation. Inoculum was prepared in malt yeast extract medium containing 0.3 % yeast extract, 0.3% malt extract , 0.5% pepton and 1% dextrose at natural pH 6.8 at 55 0 C for 3 days. Preparation of solid state fermentation: 30 gm of wheat bran was taken in one litre flask containing 70% moisture at neutral pH. Moistened bran was sterilized for 15 minutes, cooled and then inoculated with 1% spore suspension and incubated at 55⁰ C for 5-7 days. Enzymes were extracted by adding 5 volume of water containing1% (w/v) NaCl solution. h the application of enzyme. After adequate stirring the container was left for 2 hours with occasional shaking. The slurry was filtered through a sieve of 100 mesh and the filtrate was centrifuged at 600 rpm. The enzyme activity in the filtrate was assayed by DNS method.

Achievement/Findings:

Bleaching trial experiment of pulp with and without enzyme. bleaching experiments were conducted in CEH sequences with and without enzyme pretreatment to reach a final brightness of 80% ISO. The CEH sequences were conducted in conventional conditions in both Soda –AQ and Kraft pulp. In chlorination stage of CEH sequence, about 8%, 9%and 12% chlorine were reduced by the application of 1 IU, 2IU and 4IU of xylanase respectively in Soda –AQ pulp.But in the Kraft pulp 5%, 7% and 14% chlorine were reduced by the same Iu charge of xylanase. In addition, enhancement of brightness was observed with the application of enzyme.
