Studies on Rice Anther Culture

Abstract/ Executive Summary:

Anther of nine rice genotypes were studied to fined out the ability of callus induction and regeneration of plantlets. Binnatoa showed the highest percentage of callus development (1.20%) followed by the IR55182 (1.15%). IR45427 produced the lowest number of calli. The frequency of regenerated plants ranged from 15% to 55%. The regenerated calli yielded both green and albino plants. It was observed that Binnatoaproduced the highest percentage of regeneration plants (55%).

Objectives:

To develop regenerated calli from anther of rice

Methodology:

Some rice lines were used to study their ability of callus induction and regeneration of plants. For anther culture, boots were collected when the collar of the flag leaf was 5-10 cm above the collar of the penultimate leaf. Panicles were treated with cold shock by sealing the boots with polyethylene bags and refrigerating at 7-10 0C for 4-7 days. After cold treatment panicles were surface sterilized with 70% alcohol. For callus induction anthers, containing pollen in the mid-late-uninucleate stage were plated in the wide mouthed 100 ml conical flasks containing sterilized semi-solid Chaleff`s R2 medium (Autoclaved at 15 Ib for 15 minutes at pH 5.8) which supplemented with 2.0 mg/I (NAA) + 0.5 mg/1 (Kinetin). Conical flasks containing anthers were kept at dark in incubation at 25+- 10 C for callus induction. Callus formation started from 35 -65 days. For regeneration of plants, 10-20 days old calli were then transferred to MS medium supplemented with 1.0 mg/I (IAA) + 1.0 mg/1 (Kinetin) and placed at 26+- 20 C with 12 hours photoperiod. Regenerated shoots were then transferred to the medium containing ½ MS + 0.5 mg/I (MET) and kept until roots developed properly. Data on induction of callus and regeneration of green and albino plants from anther calli were recorded.
