Isolation of Microorganisms Containing Hydrolases and Yeasts from Various Agro-industrial Wastes of Bangladesh and Studies Their Possible Application on Jute Processing Systems.

Abstract/ Executive Summary:

Three thermophilic xylanolytic microorganisms were isolated from self-heating piling bin of jute cuttings. These were Thermomyces lanuginosus, Penicillium sp and Streptomyces sp. Isolation and purification was carried out on PDA media by sub culturing the microbes for several times at 55oC temperature. Then the identification of these microbes was done by observing colony morphology and microscopic features. Three types of colonies were found on PDA media.

Objectives:

There are very useful applications of cellulase-free xylanase in pretreatment of low quality jute fiber, which amounts to about one third of total world jute production. The bottom part of the jute plant is not retted properly through as usual retting process due to excess amount of hemicelluloses and pectin. The spinning process needs pretreatment for softening, which requires the removal of xylan without affecting the fiber strength. So, the improvement of pretreatment process with cellulase-free xylanase which is thermostable could be attractive over conventional oil/water emulsion treatment.

Methodology:

The experiment was carried out in the laboratory of Microbiology and Biochemistry Department of Bangladesh Jute research Institute and the Department of Microbiology, University of Dhaka, Bangladesh. Samples were collected from piling of Jute where temperature raised to 60-70°C of Pubali Jute Mill, Norshingdi. Organisms were grown on PDA media at 55°C, preserved at 40C during experiment and for long term preservation organisms are kept at -200C. For solid state fermentation wheat bran medium was used .Progress of a solid-state fermentation with selected microbial isolates was carried out at different incubation periods (1-9 days) at 55°C.

Achievement/Findings:

Their ability to produce Xylanase and CMCase enzymes under solid state fermentation was also studied using wheat bran medium as a sole source of carbon and nitrogen with 50% moisture content. Synthesis of xylanase and CMC-ase commenced within 72 hours and were reached maximumm within 6 days. Thermomyces lanuginosus produced cellulase free xylanase but Penicillium sp and Streptomyces sp. produced both cellulase and xylanase together.
