Production and Optimization of Xylanase and Cellulase by Using the Identified Wild Straints of Trichomerma Spp. in Shake Culture Fermentation.

Abstract/ Executive Summary:

Cellulase yield by the fungus Trichoderma viridae was relatively inferior as compared to strains of T. reesei. The highest enzyme activity was estimated by xylanase 13.25U/ml in comparison CMCase (1.3U/ml) and FPase (0.11U/ml).

Objectives:

To produce lignocellulolytic enzymes from Trichoderma spp. and their characterization in deferent condition (pH, temperature and agitation) and unmask their potential uses in jute products.

Methodology:

Cellulolytic fungus Trichoderma viridae was used in this experiment. Potato dextrose agar (PDA) medium was used for growth and characterization of the isolated fungus. Mendel’s (modified slightly) medium was used for enzyme production from the fungal mass. Carboxymethylcellulase (CMCase) and Xylanase were isolated from Trichoderma viridae. Carboxymethylcellulase activity was estimated according to IUPAC instruction using 1% solution of carboxymethyl cellulose-sodium salt in 0.05M citrate buffer (pH 5.0). Filter paper (Fpase) activity was determined also according to Ghose (1987) and xylanase activity was determined according to Biley et al. (1992) using 1% birch wood xylan powder (in 0.05 M citrate buffer, pH 5.0) as substrate. The liberated reducing sugars were estimated by the DNS method.

Achievement/Findings:

In this experiment highest endoglucanase (CMCase) activity was found to be 1.300U/ml, FPase activity was 0.110U/ml and xylanase activity was 13.25U/ml. Variation of enzyme activity depends on various factors. Cellulase is moderately inhibited by glucose and strongly inhibited by cellobiose. For both sugars the extent of inhibition increases with increasing resistance of the cellulose. Cellobiose is competitively inhibited by glucose.
