Improvement of Mature Embryo-based in Vitro Regeneration System in Indica Rice Cultivars for High Frequency Plantlets Production

Abstract/ Executive Summary:

The experiment was conducted during March to September 2010 at the Tissue Culture Laboratory of Biotechnology Division, Bangladesh Rice Research Institute (BRRI), Joydebpur, Gazipur to investigate the effects of medium and plant growth regulator on callus induction and plantlet regeneration in indica rice varieties. BRRI dhan48 produced the highest percentage 59.60% of callus induction. BRRI dhan28 was also produced vigorous, whitish and healthy callus. BRRI dhan49 produced the lowest percentage 26.43% callus were very poor, reddish with dead tissue.Kn was more effective than BAP in relation to regeneration and organogenesis. BRRI dhan48 and MS medium are to be recommended for callus induction. For regeneration, 2.0 mgL^-1 NAA and 2.0 mgL^-1 Kn in MS medium are to be recommended.

Objectives:

To investigate the effects of medium and plant growth regulator on callus induction and plantlet regeneration in indica rice varieties.

Methodology:

The experiment was laid out following a Completely Randomized Design where each treatment was replicated three times. Ten varieties BR5, BR11, BRRI dhan28, BRRI dhan29, BRRI dhan33, BRRI dhan41, BRRI dhan47, BRRI dhan48, BRRI dhan49 and BRRI dhan50 were used for callus induction on three different media namely MS, N6 and LS. The best variety for callus induction on the best medium was used for plantlet regeneration in subsequent experiments. The basal medium was supplemented with 30 gL^-1 sucrose, 0.3 gL^-1 casamina and 2, 4-D 2 mgL^-1 and solidified with 7 gL^-1 agar. The pH of the medium was adjusted to 5.8 prior to autoclave at 121⁰C with 1.16 kgcm^-2 pressure for 20 minutes. The culture was incubated in growth room at 25±2 ⁰C under 12 hr. photoperiod of 2000 µmolm-2s-1 provided by florescent tubes.

Achievement/Findings:

Among the studied varieties BRRI dhan48 produced the highest percentage 59.60% of callus induction. The calli were yellowish, glassy and healthy over other varieties. BRRI dhan28 was also produced vigorous, whitish and healthy callus. BRRI dhan49 produced the lowest percentage 26.43% callus were very poor, reddish with dead tissue. Among medium MS was the best for callus induction (47.00%) followed by N6 (36.08%) and LS (33.88%). The percentage of callus induction from explants was the highest 71.55% in BRRI dhan48 culture in MS medium. The highest percent plantlet regeneration (70.00%) was obtained from 2.0 mgL^-1 NAA and 1.0 mgL^-1 BAP in MS medium. The number of plantlets explant^-1 (12.33), average plantlet height (10.18 cm), average root length (7.33 mm) and number of root plantlet-1 (5.90) were also found best from above PGR combination in MS medium. MS medium supplemented with 2.0 mgL^-1 NAA and 2.0 mgL^-1 Kn was found the best in respect of percent plantlet regeneration (76.67%) from explants, number of plantlets explant^-1 (12.17), average plantlet height (9.17 cm), average root length (9.17 mm) and number of root plantlet^-1 (10.69).
