Germination Performance of Blackgram Under Long Term Conservation in the Gene Bank of Pgrc

Abstract/ Executive Summary:

Germination performance of black gram (Vigna mungo (L.) Hepper) seeds was studied on 36 accessions after 21 years from base collection at -18⁰C of Plant Genetic Resources Centre. The germination test was conducted on filter paper at 24 to 32⁰C at dark for one week. The germination test was conducted with the same accessions on soil in green house at 30 to 40⁰C and also field conditions for two weeks. The maximum germination percent was found at the Laboratory conditions (98%), followed by green house (84%) and the minimum germination percent was observed in field conditions (77%). The critical germination percentage (≥80%) was in 97%, 81% and 56% accessions from laboratory, green house and field, respectively. Normal seedlings were produced from the seeds and the plants produced normal fruits and seeds in the green house and field conditions.

Methodology:

The initial germination percentage of black gram was more then 98% before keeping it in the gene bank. Seeds were dried to 5–8% moisture content and then hermetically stored at –18⁰C. The seeds were dried and then kept it some days in silica gel. The seeds were transferred into medium term storage (4-6⁰C) when electricity and mechanical problem. Monitored seeds of black gram, had been stored in the gene bank for 21 years (since 1991). The accessions were tested on filter paper and in dark at the Seed Health Testing Laboratory of PGRC during 13-25 February 2012. Seeds were germinated at a temperature of 24 to 32⁰C for 7 days. To avoid the depletion of accessions in storage the monitored amounts of each accession was reduced to one-fifth the standards. Data were recorded on 3 to 7 days. The same accessions were tested at the experimental field of PGRC containing silty clay soil and a pH of 6.5. The fertilizer doses such as 10 ton/ha cow dung, 45 kg urea, 95 kg TSP and 40 Kg MP/ha were applied in the field. The seeds were planted in 3 meter long of two rows plot. Date of seed planting was 4 March 2012. Seedlings were counted on 10 days after seed emergence. The same accessions were tested in nine litre capacity of plastic pots. The pot was filled up with loamy soil. Cowdung of 10 kg, urea 45 kg, TSP 95 kg and MP 40 kg/ha were applied in the soil. The plastic pot was placed in the green house at 30 to 40⁰C. Date of seed planting was 13 March 2012. Seedlings were counted on 10 days after seed emergence. Number of normal seedling, abnormal seedling and dead seeds were counted and viability (%) was calculated.
