Isolation of Microbes from Different Natural Environmental Sources (chicken Soup, Onion, Guava, Cucumber, Kul, Retting Effluent from Kishoregonj and Monirampur Etc.)and Studies of Their Retting Properties

Abstract/ Executive Summary:

The experiment was undertaken to isolate microbes (bacteria & fungi) from different natural sources. The experiment was carried out in the laboratory of Fibre Quality Improvement Division during the period of 2012-2013. First of all, different natural sources containing unknown microbes were collected and those are analyzed in a sterile condition for hunting newer active jute retting microbes for retting purpose within a short period. There were 10 different fungi isolated from different natural sources and 3 fungi e.g. penicillium sp., A. niger and spectotrix sp of them were found good retter which retted green jute ribbon within 11, 11 and 12 days respectively. Four fungi e.g. Aspergillus sp., Botryodiplodia sp., Mucor sp., Fusarium sp. of them were found retter which retted green ribbon within 13, 14, 14 and 15 days respectively. The rest three fungi namely Oidium sp. Pleurotus sp. & Trichoderma sp. were found non retter.

Objectives:

i) To isolate microbes (bacteria & fungi) from different natural environmental sources

ii) To determine the retting properties of microbes on jute and allied fibre (JAF) plants.

Methodology:

Various strains of microbes were collected from different natural and environmental sources such as chicken soup, Retting effluent of Kishoreganj, Faridpur, Monirampur, onion, guava, cucumber, palm (green), kul/ boroi and pine apple which were collected from different places under this study. Collected microbes were grown in PDA, nutrient broth, peptone agar and other selective synthetic media at room temperature using standard microbiological methods and the microbes were then identified. The deshi and tossa jute varieties viz. CVL-1 and O-9897 were grown in different jute research stations and then green jute stems were brought to the Laboratory of Fibre Quality Improvement Division. The stems were cut into 8 inches pieces and the identified microorganisms were applied on the pieces of green jute stem and their retting properties were assessed.

Achievement/Findings:

In 2012-2013 year, ten different fungi were isolated from various natural sources such as chicken soup, onion, guava, cucumber, retting effluent of Kishoreganj, Faridpur and Monirampur station, palm fruit, Kul and Pine apple etc. Among the 10 isolated fungi, 3 fungi were found good retter and they are penicillium sp., A. niger and spectotrix sp. which retted green jute ribbon within 11, 11 and 12 days respectively. Four fungi were found retter namely Aspergillus sp., Botryodiplodia sp., Mucor sp., Fusarium sp., which retted green jute ribbon within 13, 14, 14 and 15 days respectively. The rest three fungi namely Oidium sp. Pleurotus sp. & Trichoderma sp. were found non retter. From the results, it is concluded that isolated good retting fungi were preserved in laboratory which may be used for retting the jute effectively in future.
