Effect of Gamma Rays on Callus Induction and Regeneration of Shoots in Salt Tolerant Rice and High Yielding Variety

Abstract/ Executive Summary:

Different doses (0, 3,4,5,6 7 and 8 Gy) of gamma ray were treated calli of Binnatoa, Pokkali and BR 27. Calli with gamma rays showed the survival percentage decreased with the increased in radiation doses. Shoots were regenerated from all the doses except 8 Gy in Binnatoa, 5 Gy in Pokkali and BR 27. After irradiating calli, LD-50 values were calculated from the regeneration shoots and it was found to be 5 Gy in Binnatoa and 2 Gy Pokkali and BR 27.

Objectives:

To develop callus and shoots from induced gamma rays of salt tolerant rice

Methodology:

Three to four week old calli obtained from varieties viz. Binnatoa, Pokkali, and BR 27 were irradiated with different doses of gamma rays (0,1, 2, 3, 4, 6, 7 and 8) to create variability’s in the regeneration medium and the medium was MS supplemented with 0.5 mg/1 NAA and 11.0 mg/1 Kinetin. Survival percentage of calli and frequency of regenerated shoots were recorded.

Achievement/Findings:

After irradiatng calli, LD₅₀ values were caculated from the results on regeneration of shoots and it was found to be 5 Gy in Binnatoa and 2 Gy in Pokkali and BR27.
