Influence of Tempareture and Colour of Polythylene Bags on Shelf-life of Mass Cultured Trichoderma Isolates at Storage Condition

Abstract/ Executive Summary:

In-vitro and in-vivo biological control of certain pathogens are greatly concern to the selection of effective Trichoderma isolate and minimum level of shelf life of the mass produced inocula prevailing on stored at the normal freezing (4⁰C) temperature than room temperature.Shelf life of mass cultured Trichoderma blue colored bags might be selected for long term storage and another findings is to screen an effective Trichoderma isolates.

Objectives:

To test the viability termed as colony forming unit(cfu) using PDA media in laboratory condition.

Methodology:

Three isolates were finally selected for mass multiplication with some organic matters as well as it was preserved in the soil of marigold nursery bed for future isolation as natural culturing strategy. The organic matters are Cow Dung (CD), Poultry Refuse (PR), Sand (S) and Maize Meal (MM) was mixed for colonization of Trichoderma strains. The ratio was CD: SD: PR: MM = 8: 4: 2: 1. Data were taken of the Trichoderma strains colonized organic substrate(1g mixed with 100ml sterilized water) in the room and freezing (4⁰C ) temperature 210 days after inoculation with 30 days of interval.. Transparent, semitransparent, blue and dark (black) colored polyethylene bags were used to store the Trichoderma inocula for 180 days.

Achievement/Findings:

The Strain-2 (A03F2005) of Trichoderma gave highest cfu both in room and freezing temp. up-to 210 days after inoculation. The initial no of spore germination was 3.5X108 and after 210 days after inoculation, satisfactory no. of cfu was harvested (1.3X104 , 1.3X105) while the Trichoderma Strain-1 ((A03D2004)) and Trichoderma Strain-3 (B07D2005) showed poor number of shelf-life viability after same days of storage both in room and freezing conditions. Form the result it is concluded that the successful in-vitro and in-vivo biological control of certain pathogens are greatly concern to the selection of effective Trichoderma isolate and minimum level of shelf life of the mass produced inocula prevailing on stored at the normal freezing (4⁰C) temperature than room temperature. Blue colored poly bags gave highest colony forming unit at 180 days after inoculation in case of three strains of Trichoderma followed by semitransparent and transparent poly bags. The lower no. of cfu was obtained from the dark poly bag. Among the Trichoderma isolates, the strain-2 showed highest performance to have shelf-life viability for long 180 days after inoculation. Blue bag produce 1.0x106 colony followed by transparent poly bag (4.8X105), Semi-transparent poly bag (3.2X104) and dark poly bag (2.5x103)/g sample. Strain-3 (B07D2005) failed to produce any viable spore 180days after inoculation. It is concluded from this experiment that for adequate shelf life of mass cultured Trichoderma blue colored bags might be selected for long term storage and another findings is to screen an effective Trichoderma isolates.
