Acclimatization of Micro Propagated Potato Plantlets from in Vitro to Ex Vitro Conditions

Abstract/ Executive Summary:

The experiment was conducted at the Tissue Culture Laboratory, TCRC, BARI, during 2007-08 potato season. The design was CRD two factors with three replications. Two sizes of pots and three media components were used in the experiment.The pot size had no effect on plantlet establishment. The media components had the significant effect on plantlet establishment. The media, M₄(1/4 Sandy soil + 1/4burnt rice husk +1/4 decompost cowdung) in medium size pot produced the maximum number (3.26) of tuber and weight (38.47 g) per plantlet.

Objectives:

To optimize the most suitable pots and media for hardening of plantlets.

Methodology:

The experiment was conducted at the Tissue Culture Laboratory, TCRC, BARI, during 2007-08 potato season. The design was CRD two factors with three replications. Two sizes of pots P₁=Medium 8.0 cm (height) × 9.0 cm (top diameter) × 6.4 cm (bottom diameter) and P₂= small size= 3.4 cm (height) × 3.3 cm (top diameter) × 1.9 cm (bottom diameter) and five media components were used for hardening of plantlets. The media components were M₁=1/2 Sandy soil +1/2 Decomposed cowdung, M₂= 1/3 Sandy soil + 1/3 Coconut dust + 1/3 Decompost cowdung, M₃= 1/3 Sandy soil + 1/3 burnt rice husk + 1/3 decompost cowdung, M₄= ¼Sandy soil +¼ Coconut dust + ¼ burnt rice husk +1/4 Decompost cowdung and, M₅= 1/5 Sandy soil + 1/5 Coconut dust +1/5 burnt rice husk + 1/5 Saw dust + 1/5 decompost cowdung. All media were properly sterilized. Six plantlets were planted in each plot for each treatment. The plantlets were transferred to pots for hardening after 3 weeks of inoculation in the test tube. The potted plants were kept in a shed condition. The plantlets were transferred from pot to field in the net house after 7 days of planting. Irrigation was applied morning and evening with watering can in a fine mist.

Achievement/Findings:

There were no significant differences between the pot sizes for plantlet establishment, tuber number and weight. The small pot with media component, M₄= Sandy soil + Coconut dust + burnt rice husk + decomposed cow dung (1: 1: 1:1v/v) is most effective and economic for plantlet hardening.
